Multi-Residue Analytical Method for Antibiotics

All standard chemicals used for analyses were purchased from Dr. Ehrenstorfer GmbH (Augsburg, Germany), except for the internal standards (erythromycin-13C-d3, lincomycin-d3, sulfamethoxazole-d4, ciprofloxacin-d8, doxycycline-d3, and chloramphenicol-d5), which were purchased from Sigma-Aldrich (St. Louis, MO, USA). High performance liquid chromatography (HPLC)-grade solvents, including methanol, formic acid, and acetonitrile, were provided by Merck & Co., Inc. (Kenilworth, NJ, USA). Guaranteed reagents, including ammonium acetate, hydrochloric acid, sodium hydroxide, dipotassium hydrogen phosphate, and disodium ethylenediaminetetraacetic acid (EDTA), were purchased from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). Ultrapure water was prepared using a water purification system. The internal standards were dissolved in acetonitrile as stock solutions and stored in a freezer at −40 °C. Working standard solutions were prepared immediately before measurements were taken by diluting the stock solutions with a mixture of menthol and water (9:1, v/v). For solid-phase extraction (SPE), Oasis HLB cartridges (6 mL/500 mg) were purchased from Water Oasis Co., Ltd. (Milford, MA, USA), glass microfiber filters (Whatman GF/F) were obtained from Sigma-Aldrich (St. Louis, MO, USA), and syringe-driven filters (PTFE) from Millipore Corp. (Bedford, MA, USA).

MilliQ water from a MilliQ system (Millipore, Bedford, USA) was used throughout this study.

Analytical Quantification of Sulfamethoxazole

The cartridges used for solid phase extraction, OASIS HLB (200 mg, 6 mL), were obtained from Water Corporation (Milford, MA, U.S.A.). HPLC grade methanol and water were purchased from Merck (Darmstadt, Germany). Sulfamethoxazole standard as well as the isotopically-labelled internal standard sulfamethoxazole-d4 were purchased from Sigma-Aldrich (St Louis, MO, USA). Sulfamethoxazole metabolites, N-acetyl sulfamethoxazole, desamino-sulfamethoxazole and glucoronide sulfamethoxazole were purchased from Toronto Research Chemicals (TRC) (Ontario, Canada). All analytical standards were HPLC suitable and were prepared in methanol at a concentration of 1000 mg/L and stored at -20 ºC. Working standard solutions (1 mg/L) were prepared in methanol before each analytical run.

Analytical Method for Emerging Contaminants

Reagents purchased were HPLC grade and the standards used were of high purity grade (>95%): perfluorooctanoic acid (PFOA), perfluoroheptanoic acid (PFHpA), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluoroundecanoic acid (PFUnDA), bisphenol A (BPA) and acetaminophen (ACT), caffeine (CAF), diclofenac (DCF), lamivudine (LA), triclosan (TS), phenytoin (PHE), sulfamethoxazole (SMX), carbamazepine (CAR), 2nitrophenol, sulfamethoxazole-d 4 and acetaminophen-d 4 from Sigma Aldrich (South Africa). Individual stock standards and isotopically labelled internal standards were prepared in methanol at a concentration of 1000 mg/L. Stock solutions in methanol were stored at À20 C. Working standard solutions were prepared by appropriate dilution of the stock solutions in methanol stored at 4 C in appropriate storage amber container glassware to prevent light degradation. Working standard solutions were used for preparation of the calibration curves and for spiking samples in the validation study. The cartridges Oasis HLB (6 mL, 200 mg) were obtained by Waters Corporation (Mildford, MA, USA). HPLC grade methanol, acetonitrile and acetone from Sigma Aldrich (South Africa), ultrapure water was purified by Milli-Q system (Millipore, Bedford, MA, USA).
