Cell Viability Assay with GNPs

Check if the same lab product or an alternative is used in the 5 most similar protocols
The CCK8 assays were performed as instructed by the manufacturer to assess cell viability. Briefly, the cells were seeded in 96-well plates (approximately 3000 cells/well). GNPs and GAL-PEG-GNPs were diluted to various concentrations in 1× PBS (pH 7.4), then added into the wells. After 24 h incubation, 20 μL CCK8 (keygentec company, Nanjing, Jiangsu, China) was added to each well for 4 h. Optical density (OD) was measured at 490 nm using a Microplate reader (BioRad, DG3022, USA) and the proliferation index was calculated as experimental OD value/control OD value. Estimate value of 50 % inhibition concentration (IC50).

Cytotoxicity Evaluation of C225, GNPs, and C225-GNPs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cell Counting Kit-8 (CCK8; Nanjing KeyGen Biotech Co., Ltd.) assays were used to determine the cytotoxicity of C225, GNPs and C225-GNPs. The assays were performed according to the manufacturer's protocol to assess cell viability. The cells were cultured at a density of 3×103 cells per well in flat-bottomed 96-well plates. The C225, GNPs and C225-GNPs were diluted to various concentrations in 1X PBS (Ph 7.4), and then added into the wells and incubated for 24 h at 37°C, followed by exposure to 10 µl CCK8, which was added to each well for 2 h. The absorbance was measured at 490 nm using a microplate reader (DG3022; Bio-Rad Laboratories, Inc., Hercules, CA, USA) and the 50% inhibition concentration (IC50) value was estimated.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
