Mesangial Cell Glucose Exposure Protocol

Rat glomerular mesangial cells (HBZY-1) were obtained from Boster (Wuhan, China, Product code: CX0130). The cells were incubated at 37 °C, under a 5% CO2 atmosphere and cultured in DMEM containing 10% FBS. Cells in passage 6–12 were used in the experiments. To determine the biological activity of AC and marein in vitro, HBZY-1 cells were starved in serum-free medium for 12 h. Normal control (NC) cells were cultured in DMEM medium containing 5.5 mM glucose. The model cells were treated/or not with different concentrations of AC and marein for 2 h, followed by exposure to 50 mM glucose in DMEM medium for 24 h [24 (link)].

TLR4 Overexpression in Rat Mesangial Cells

Rat glomerular mesangial cell line (HBZY-1) was obtained from Boster Biological Technology Co., Ltd. (Wuhan, China) and maintained in Dulbecco’s modified Eagle’s medium (DMEM) containing 10% fetal bovine serum (FBS) at 37°C. The TLR4 overexpression plasmid pcDNA3.1-TLR4 and empty pcDNA3.1 vector were purchased from GenePharma (Shanghai, China). Transfection of pcDNA3.1-TLR4 and pcDNA3.1 vector was performed using Lipofectamine 2000 (Invitrogen, Carlsbad, CA, United States) according to the manufacturer’s instructions. At 48 h after transfection, western blot was performed to determine transfection efficiency.
