Cell counting kit 8 cck 8

Manufactured by US Everbright
Sourced in China

The Cell Counting Kit-8 (CCK-8) is a colorimetric assay for the determination of cell viability and proliferation. It utilizes a water-soluble tetrazolium salt that is reduced by cellular dehydrogenases, producing a yellow-colored formazan dye. The amount of this dye is directly proportional to the number of living cells, making it a reliable method for quantifying cell proliferation and cytotoxicity.

Adenovirus and siRNA Treatment on Cell Proliferation

Check if the same lab product or an alternative is used in the 5 most similar protocols
GCS were seeded onto 96-well plates at 1 × 104/well and treated with recombinant adenovirus and siRNA for 0 h, 12 h, 24 h, 36 h and 48 h, separately. Cell proliferation was detected with Cell Counting Kit-8 (CCK8) (US Everbright® Inc., Nanjing, China). According to the instructions of the manufacturer, the cells were added to 10 μL of CCK8 solution and incubated at 37 °C for 2 h. Then, the optical density (OD) value at a wavelength of 450 nm was detected by a microplate reader (Bio-Rad, Hercules, CA, USA).

Melatonin and 740 Y-P Neuroprotection

Check if the same lab product or an alternative is used in the 5 most similar protocols
Melatonin (HY-B0075) and 740 Y-P (HY-P0175) were bought from MedChemExpress (MCE, China). A 1mol/L stock solution was attained by dissolving melatonin in dimethyl sulfoxide (DMSO), which was then kept at −20°C in the dark. The DMSO and N-acetyl-L-cysteine (NAC) were provided by the Beyotime (Beyotime Institute of Biotechnology, Shanghai, China). The Cell Counting Kit-8 (CCK-8) was bought from US EVERBRIGHT INC. The primary antibodies used in this study included Bax (ab32503, Abcam, Cambridge, MA, USA), Bcl-2 (ab32124, Abcam, Cambridge, MA, USA), Cytochrome C (ab76237, Abcam, Cambridge, MA, USA), Cleaved Caspase-3 (9662, Cell Signaling Technology Inc, CST, MA, USA), PI3K (AB3263, Technology, Shanghai, China), phospho-PI3K (CY6427), phospho-Akt (AY0421), and Phospho-mTOR (CY5996). The HRP-linked goat anti-mouse and anti-rabbit secondary antibodies were also bought from CST.

Cellular Proliferation Assays of Thyroid Cancer Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cell proliferation assays were performed using Cell Counting Kit-8 (CCK-8; C6005M; US Everbright; Silicon Valley, CA, USA) and 5-ethynyl-2′-deoxyuridine (EdU; C6015M; US Everbright) kits according to their manufacturer’s instructions. For the cell viability assay, PTC cells were inoculated into 96-well plates at a density of 1,000 cells/well and left to stand with 10 μL of CCK-8 reagent for 2.5 h at 37°C with 5% CO2, then incubated for 0, 24, 48, 72 and 96 h following transfection. For the EdU assay, 5 × 104 cells were inoculated into a 24-well plate and cultured in complete culture medium. After 24 h, cells were stained and photographed according to the manufacturer’s instructions. For the colony formation assay, the corresponding siRNA was transfected into TPC-1 and KTC-1 cells for 24 h. Then, cells were digested with trypsin and transferred into six-well plates (1000 cells/well) and kept for two weeks for colony formation assays. All experiments were repeated at least three times.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
