Efficient mESC Transfection Protocol

mESCs were trypsinized and washed with 1× PBS (Nacalai Tesque) once. Cells (2 × 105) were transfected with 40 pmol of siRNA and 20 μl of the P3 Primary Cell Nucleofector Solution (Lonza; supplement 1 added) using program CG-104 in 96-well Shuttle Device (Lonza) according to the manufacturers’ instructions.
