Isolation and Expansion of Human CD34+ Cells

Citric acid (ACS reagent, ≥99.5%), Diethylentriamine (DETA, 99%), L-Glutamine-Penicillin-Streptomycin solution, Dulbecco’s Phosphate Buffered Saline (DPBS), Float-A-Lyzer dialysis devices (100–500 Da), human serum albumin, EDTA, Selenoprotein-AF647 antibodies and sterile filters (200 nm) were obtained from VWR and antibodies against CD45-FITC/CD34-PE, CD34-APC and the FITC Annexin V Apoptosis Detection Kit I were purchased from BD biosciences. Stem SPAN™ SFEM II medium, Stem SPAN™ CD34+ Expansion Supplement (10x) and Lymphoprep™ solution were bought at STEMCELL™ Technologies and microwave reaction vessels were obtained from CEM GmbH. The CD34 MicroBead Kit UltraPure human, MACS LS columns and 30 pre separation filters were purchased from Miltenyi Biotec and the Fix and Perm Kit was bought from Thermo Fisher Scientific. Separation buffer was prepared freshly by supplementing 500 ml DPBS with 1.5 ml 5% human serum albumin and 1.5 ml 50 mM EDTA.

Biotinylated Aptamer Surface Preparation

Biotin PEG thiol (MW 1000 Da) was purchased from Nanocs. PEG thiol (MW 550 Da) was purchased from Creative PEGWorks. Neutravidin (NA) and tris(2-carboxyethyl)phosphine hydrochloride (TCEP) were purchased from Thermo Fisher Scientific. Polystyrene beads (0.6 µm mean particle size), sulfuric acid (95.0–98.0%), hydrogen peroxide (30% w/w in H2O), and ethyl alcohol (200 proof) were purchased from Sigma-Aldrich. Paraformaldehyde (16% w/v aq soln, methanol free), Dulbecco’s Phosphate Buffered Saline (1×, without calcium and magnesium), and micro cover glass (No. 1, 18 mm × 18 mm) were purchased from VWR. Lysogeny broth and granulated agar were purchased from BD Difco. Biotinylated aptamer (Bt-aptamer, 5′[Bt]-ATACCA-GCTTATTCAATTCCCCCGTTGCTTTCGCTTTTCCTT-TCGCTTTTGTTCGTTTCGTCCCTGCTTCCTTTCTTG-AGATAGTAAGTGCAATCT3′) and fluorescently labeled biotinylated aptamer (6FAM-aptamer-Bt, 5′[6-carboxyfluorescein-ATACCAGCTTATTCAATTCCCCCGTTGCTT-TCGCTTTTCCTTTCGCTTTTGTTCGTTTCGTCCCTG-CTTCCTTTCTTGAGATAGTAAGTGCAATCT-[Bt]3′) were synthesized by Sigma-Aldrich. Deionized (DI) water used in all experiments was prepared using a Milli-Q Gradient water purification system with a resistivity of 18.2 MΩ·cm at 25 °C (Millipore).

Isolation of Brain Microvessels

After euthanasia, the meninges were removed and cortices were dissected in cold Dulbecco's phosphate‐buffered saline (02–0119‐0500; VWR life science, USA). Cortices were homogenized using a Dounce grinder and centrifuged (4°C) for 5 min at 2000 g. The vessel pellet was resuspended in 15% 70‐kDa dextran (31,390, Sigma‐Aldrich, USA) solution and centrifuged (4°C) for 15 min at 10,000 g. The upper layer, containing myelin and brain parenchymal cells, was removed. Vessels were retrieved by pipetting and transferred to a 40‐μm cell strainer. After being washed with cold DPBS (14,190,144; Gibco, USA), the vessels were collected in a tube by inverting the filter and adding 10 mL of MCDB 131 medium (10372–019; Gibco, USA) containing 0.5% fatty acid‐ and protease‐free BSA (16,609; Sigma‐Aldrich, USA). The suspension can be centrifuged (4°C) for 1dain at 5000 g to pellet the vessels for further study.

In Vitro Metabolism of Cannabinoids

Pooled human liver microsomes (HLMs) [n = 50, mixed gender (21 female and 29 male), race (42 Caucasian, 4 Hispanic, 2 African American, and 2 Asian), and age (5–77 years)] and pooled human kidney microsomes (HKMs) [n = 8, mixed gender (50% each), race (3 African American, 3 Caucasian, and 2 Hispanic), and age (42–70 years)] were obtained from Sekisui Xenotech, LLC (Lenexa, KS). β-estradiol, chenodeoxycholic acid, trifluoperazine, serotonin, propofol, codeine, zidovudine (AZT), nicotine, oxazepam, dihydroexemestane, ketoconazole, diclofenac, acetaminophen, and furosemide were all purchased from Sigma-Aldrich. Optima grade methanol, acetonitrile, and formic acid were obtained from Fisher Scientific (Waltham, MA). Ultra-low-binding microcentrifuge tubes, Dulbecco’s modified Eagle’s medium, Dulbecco’s phosphate-buffered saline, UDPGA, alamethacin, MgCl2, and geneticin (G418) were purchased from VWR (Radnor, PA). BCA protein assays were purchased from Pierce (Rockford, IL), premium grade FBS was purchased from Seradigm (Radnor, PA), and ChromatoPur bovine serum albumin (BSA) was purchased from MB Biomedicals (Santa Ana, CA).

Quantitative Analysis of CBD and 7-OH-CBD Metabolism

were obtained from Sekisui Xenotech, LLC (Lenexa, Kansas). NADPH-regenerating
system (1.3 mM NADP, 3.3 mM glucose 6-phosphate, and 0.4 U/mL glucose
6-phosphate dehydrogenase) was obtained from Corning (Bedford, MA).
Nicotine, norNicotine, HPBA, NOX, Nic-Gluc, cotinine, COX, Cot-Gluc,
3HC, and 3HC glucuronide (3HC-Gluc) standards were purchased from
Sigma-Aldrich. Fluconazole, benzydamine hydrochloride, clopidogrel,
trifluoperazine, diclofenac, amitriptyline, and tranylcypromine were
also purchased from Sigma-Aldrich. Optima grade methanol, acetonitrile,
and formic acid were obtained from Fisher Scientific (Waltham, MA).
Ultra-low-binding microcentrifuge tubes, Dulbecco’s modified
Eagle’s medium, Dulbecco’s phosphate-buffered saline,
UDP glucuronic acid (UDPGA), alamethicin, MgCl2, and Geneticin
(G418) were purchased from VWR (Radnor, PA). BCA protein assays were
purchased from Pierce (Rockford, IL); premium-grade fetal bovine serum
(FBS) was purchased from Seradigm (Radnor, PA), and ChromatoPur bovine
serum albumin (BSA) was purchased from MB Biomedicals (Santa Ana,
CA).
