Broth Microdilution Antibiotic Testing

MIC plates were made daily using the broth microdilution method with the standard 2-fold dilution series [81 (link)]. LB was used as the growth medium for all experiments (1% tryptone, 0.5% yeast extract, 1% NaCl). Antibiotics tested include piperacillin sodium, tobramycin, and ciprofloxacin HCl (Sigma). Aliquots of 1 mg/ml and 10 mg/ml antibiotic stocks were made by diluting the antibiotic powders in LB and were stored at −20°C. New frozen drug aliquots were used on a daily basis.

Early Human Tissue Procurement

Placenta and fetal liver were de-identified, discarded material obtained from elective termination of first and second trimester pregnancies following informed consent. As these tissues are discarded material with no personal identifiers, this research does not constitute research on Human Subjects. This protocol was reviewed by the UCLA IRB committee, who determined such studies can be performed without further IRB review. Specimen age for this study is denoted as developmental age, two weeks less than gestational age, and was determined by ultrasound or estimated by the date of the last menstrual period. Tissues were harvested into PBS 5% FBS (Hyclone), Ciprofloxacin HCl (10 ng/mL, Sigma), amphotericin B (2.50 μg/mL, Invitrogen), 1% penicillin/streptomycin, transported on ice and processed the same day.

Nephrite-Based Drug Delivery System

Nephrite was obtained from Sujenara Co. (Seoul, Korea). Dopamine, ciprofloxacin HCl (CIP), levofloxacin (LEVO), ethylene glycol dimethacrylate (EGDMA), azobisisobutyronitrile (AIBN), 2-hydroxyethyl methacrylate (2-HEMA) and tryptic soy broth (TSB) were purchased from Sigma-Aldrich Chem. Co. Ltd. (St. Louis, MO, USA). Dialysis membranes with 500~1000 g/mol and 8000 g/mol of molecular weight cut-off (MWCO) were purchased from Spectrum Lab., Inc. (New Brunswick, NJ, USA). All of the organic solvents, such as dichloromethane (DCM), dimethylsulfoxide (DMSO) and ethyl alcohol (EtOH), were used as an ultrapure grade.

Antibiotic Susceptibility Profiling of Uropathogenic E. coli and P. aeruginosa

E.coli UTI89 and E.coli CFT073 were obtained from Harry Mobley (University of Michigan); both strains have been sequenced. E.coli UTI89 is a prototypic cystitis isolate (1 (link)) and E. coli CFT073 is a prototypical UPEC isolate cultured from blood and urine of a patient with pyelonephritis (10 (link)). P.aeruginosa-Boston 41501 was purchased from American Type Culture Collection (Manassas, VA), and P.aeruginosa-PA01 was obtained from Kevin McIver; only PA01 has been sequenced. Antibiotics (Ampicillin, Ciprofloxacin and Nitrofurantoin) were purchased from Sigma-Aldrich (St. Louis, MO) and used at the following concentration ranges: Ciprofloxacin HCl (Cipro: 200 ng/mL and 2 μg/mL), Nitrofurantoin (Nitro: 20 μg/mL and 40 μg/mL), and Ampicillin (Amp: 50 μg/mL). DMSO was used as a solvent to dilute all the antibiotics. The final concentration of DMSO was 0.1%. Biofilm formation was not affected at this low concentration of DMSO.

A paste of MWCNTs (70% w/w) in mineral oil (Fluka, USA) was added to copper electrode surface. Polyvinyl chloride (PVC) of relatively high molecular mass purchased from Fluka, Germany. Milk products were purchased from a local Egyptian supermarket. Buffers of 3–10 pH range were prepared following Britton and Robinson method [41 (link)].
