Cells obtained from murine vaginal lavages of healthy mice were obtained and counted using a Cellometer K2 cell counter (Nexcelom, Lawrence, MA) to obtain quantitative values for observed leukocytes and cornified epithelial cells. Standard solutions containing 1×10⁶ cells for each condition were prepared in PBS. Additionally, OVCAR-8-RFP cells were trypsinized, resuspended in medium and subsequently counted using a Cellometer K2 cell counter. Spiked lavage samples containing 0%, 0.1%, 1%, 10% and 100% OVCAR-8-RFP cells were prepared in PBS with a final volume of 500 μL. Flow cytometry was used to measure the percentage of OVCAR-8-RFP (Cytoflex, Brea, CA). Samples comprised of 100% murine lavage cells (negative control) and 100% OVCAR-8-RFP cells (positive control) were used to set up the gates.
