Tissue DNA and RNA Extraction and cDNA Synthesis

DNA and mRNA extraction were performed on frozen tumor samples using Maxwell RSC Tissue DNA and Maxwell RSC simplyRNA Tissue kits, respectively, (Promega, USA) according to the manufacturer's protocol. The quality and quantity of the DNA and mRNA were determined using a NanoDrop ND-1000 spectrophotometer (Thermo Fisher Scientific, NanoDrop Technologies, USA). First-strand cDNA were synthesized from 1 μg of total RNA using a High-Capacity cDNA Archive Kit (Applied-Biosystems, Life technologies, USA) according to the manufacturer's protocol. The obtained cDNA samples were diluted 6 times for quantitative polymerase chain reaction (qPCR) assay.
