Sorted cTFH were cocultured with sorted autologous RM, AM, or TLM cells (2 × 104) at 1:1 ratio with SEB (1 μg/mL, Toxin Technology) in RPMI (Gibco, Thermo Fisher Scientific) supplemented with 10% FCS, 100 IU/mL penicillin, 100 mg/mL streptomycin (Life Technologies, Thermo Fisher Scientific), 1 M HEPES buffer (Corning), and 2 mM l-glutamine. Some cocultures were supplemented with mouse IL-21R–Fc (R&D Systems, Bio-Techne) or isotype-matched control (R&D Systems, Bio-Techne). After 6 days of coculture, cells were stained with CD4, CD19, CD27, CD38, and CD71 antibodies before acquisition on the cytometer. DSAs were detected in supernatants after 6 days of coculture using Luminex SAB assay.

Flow Cytometric Analysis of Lymphocytes

Lymphocytes harvested from spleen and other organs were stained and analyzed by flow cytometry. All antibodies were purchased from eBiosciences (San Diego, CA) and BD Biosciences (San Jose, CA), except anti-Granzyme B PE (Invitrogen, NY), BATF (Cell signaling, MA) and Mouse IL-21 R Fc (R&D Systems, MN). Intracellular staining of transcriptional factors and BATF were performed with Foxp3 nuclear staining protocol (eBioscience). All flow cytometry data were acquired on an LSRII (BD Biosciences) and analyzed by FlowJo (Treestar).
