Computerized Rat Tail-Cuff Blood Pressure

Blood pressure was measured by a computerized rat occlusion tail-cuff blood pressure device BP-2010A (Softron Biotechnology, Beijing, China). Briefly, rats were placed into a restraint box over a heating pad (TMC-213) preheated to 37 °C. Then, the occlusion tail cuff was connected to the sensor (BP98-RCP-M, Softron Biotechnology). After 5 minutes of acclimatization, the systolic blood pressure, diastolic blood pressure, and mean arterial pressure of the rats were recorded. The measurements were repeated 3 times.

Tail-Cuff Blood Pressure Measurement

Blood pressure measurements were performed from 8:00 a.m. to 6:00 p.m. in a warm and quiet room once a week. A rat was selected randomly for BP measurement from group A, followed by B, C, D, and E, sequentially. After one round, repeat the abovementioned operations until all BP measurements are completed. Systolic blood pressure and diastolic blood pressure (DBP) of conscious rats were measured by the tail-cuff system (BP-2010A, Softron, China) as described previously (19 (link)). Five BP measurements were recorded consecutively for each rat. If the difference between any two of the five BP readings per individual rat exceeded 10 mmHg, a sixth BP measurement was conducted. The mean value was calculated for analysis. Hypertension was defined as a SBP ≥ 140 mmHg.

Renal Fibrosis Modulation by SIRT3 in Angiotensin II-Induced Mice

129 wild-type (WT) male mice of 8 weeks old were purchased from Department of Laboratory Animal Science of Peking University (Beijing, China) and SIRT3-knockout (SIRT3-KO) mice were purchased from the Jackson Laboratories (USA). SIRT3-overexpression (SIRT3-LV) mice were obtained by injection to the 129 WT mice SIRT3-overexpression-lentivirus (JIKAI GENE, Shanghai, China) via caudal vein. HKL were intraperitoneally injected in 129 WT mice for 42 days (25 mg/Kg, once a day) as the HKL treatment group. All experimental mice anesthetized with isoflurane (1%) were implanted subcutaneously corresponding osmotic pumps beforehand filled with AngII or saline for 42 days. The osmotic pumps infused with appropriate doses of AngII in sterile saline (2000 ng/kg per min) or saline according to the directions were placed in sterile 0.9% saline at 37°C for 60 hours to prime. Together, the experimental animals were randomly assigned to eight groups: WT+saline group, WT+AngII group, SIRT3-KO+saline group, SIRT3-KO+AngII group, SIRT3-LV+saline group, SIRT3-LV+AngII group, WT+HKL+saline group, WT+HKL+AngII group. Mice were sacrificed at the end of the experiment and the kidneys were used to assay for renal fibrosis. In addition, Blood pressure were measured by Tail-Cuff way (BP2010A, softron, Japan) before modeling and death.

Noninvasive Blood Pressure Monitoring

The systolic-, diastolic- and mean blood pressures were measured at indicated weeks by a computerized noninvasive tail-cuff manometry system (BP-2010A; Softron Bio technology, Beijing, China). The five measured blood pressure values from each rat were averaged. To ensure the accuracy of blood pressure measurement, the rats were trained for one week before the experiments, in order for the rats to be acclimated to the procedure.
The rats were placed in metabolic cages for urine collection. The 24-hour-urine volumes and sodium excretions were obtained at the indicated times. The rats were acclimatized in the metabolic cages for at least 2 days before collecting the urine. Sodium concentrations in the urine samples were measured by Sodium Assay Kit (Nanjing Jiancheng Bioengineering Institute, Nanjing, China). In addition, serum selenium levels were measured by inductively coupled plasma mass spectrometry (Agilent 7900, Agilent Technologies, CA, USA).

Non-invasive Blood Pressure Monitoring in Mice

Rat Model of Cardiac Hypertrophy

This study was approved by the Ethics Committee of Xinhua Hospital. All animal experiments were performed in accordance with the National Institutes of Health Guide for the Care and Use of Laboratory Animals.
Rat CH model was generated by inducing rat with continuous infusion of Ang-II (0.7 mg kg -1 d -1 ) through a subcutaneously implanted osmotic minipump (Alzet model 2ML4, DURECT Co), as described previously. 45 (link) In brief, male Sprague Dawley rats at 7 weeks of age (180-200 g; SLAC Animal, China) were anesthetized with sodium pentobarbital (50 mg/kg IP). The minipumps were primed overnight according to the manufacturer's instruction and loaded with saline (n=10) or Ang-II (n=16) dissolved in sterile solution (0.9% NaCl and 0.01 N acetic acid). Half of the rats with Ang-II infusion (n=8) were further intragastrically administrated with Valsartan (20 mg/kg daily; Novartis, Switzerland) for 4 weeks together with the implantation. 46 (link) Rat blood pressures were measured before and after infusion of Ang-II using a sphygmomanometer for rodent (BP2010A; Softron, Japan). Four weeks after treatment, the rat hearts were harvested, weighted, snap-frozen in liquid nitrogen.
