Quantifying IL-10 Secretion by Breg Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Purified CD1dhighCD5+ Breg cells (1×105 cells/ml) were cultured in 1 ml of complete medium with or without LPI for 5 hrs. IL-10 concentration in culture supernatant was then quantified using the IL-10 OptEIA ELISA kit (BD Pharmingen, San Diego, CA, USA) following the manufacturer’s protocol. All assays were conducted in triplicate.

Cytokine Profiling of Murine and Human CLL

Check if the same lab product or an alternative is used in the 5 most similar protocols
For cytokine analysis, normal B-1, murine CLL and human CLL cells were cultured in triplicate (2×106 cells/mL) for 24 hours in 96-well plate with various stimulants or treatments. Cells were removed by centrifugation and supernatants were assayed immediately or stored at −80°C. Plasma from mice was obtained by centrifuging blood collected in EDTA tubes at the time of euthanization by cardiac puncture and the samples were stored at −80°C. IL-10 levels in supernatants or plasma were quantified using IL-10 OptEIA ELISA kit (BD#555252). IFN-γ levels were measured using IFN-γ OptEIA ELISA kit (BD#555138). Human plasma or secreted IL-10 levels were quantified using IL-10 ELISA MAX set (BioLegend #430601). Limit of IL-10 detection is 8 pg/ml.

Check if the same lab product or an alternative is used in the 5 most similar protocols
Isolated splenic CD19+ B cells (3 × 106 cells/well) and Peritoneal cavity fluid (PeC)-derived CD19+ B cells (1 × 106 cells/well) were stimulated with LPS (0, 0.1, 1, and 10 μg/ml) for 24 or 48 h, the level of IL-10 by using a mouse BD OptEIA IL-10 ELISA kit according to the manufacturer’s instructions (BD Biosciences, San Jose, CA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
