Purification and Reconstitution of AmtB

Ubiquitin (bovine erythrocytes, >90% purity) was purchased from Sigma Aldrich (St. Louis, MO). A 5.0 μM stock solution in deionized water (18 MΩ) with 0.1% formic acid was prepared and used without further purification. Ammonia transport channel (AmtB) was expressed and purified as described previously [16 (link)]. AmtB was diluted in 200 mM ammonium acetate and 0.5% C8E4. Lipid samples of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphate (POPA), 1-palmitoyl-2-oleoyl-sn-glycero-3-phospho-L-serine (POPS), 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoethanolamine (POPE), and 1′,3′-bis [1,2-dimyristoyl-sn-glycero-3-phospho]-sn-glycerol (TMCDL) were purchased from Avanti Polar Lipids Inc. (Alabama, USA). Lipids stocks were prepared in 200 mM ammonium acetate and 0.5% C8E4 and used without further purification.
