Previously transfected cells were passed into wound-healing assay culture insert (ibidi) 24 h after transfection. After 24 h of culture in the device, the insert was removed to make a wound of ∼450 µm. Images were taken at three different locations along the wound at every hour for 9 h. The distance between both sides of the wound was measured to calculate the speed of closure over the 9 h. For staining experiments, cells were fixed 6 h after removing the insert.
