Ricin Cytotoxicity Assay in Vero Cells

Vero cells were detached from culture dishes with trypsin (Gibco), seeded into white 96-well cell culture treated plates (Fisher Scientific) (100 uL per well, 5 × 104 cells/mL) and allowed to adhere overnight. The cells were then treated with Dulbecco’s Modified Eagle Medium (DMEM) alone, ricin alone (10 ng/mL), or a mixture of ricin with VHHs at five-fold dilutions. After 2 h at 37 °C, the culture medium was changed, and the cells were incubated at 37 °C for ~48 h. Viability was assessed using CellTiter-GLO (Promega, Madison, WI, USA). All treatments were performed in triplicate and repeated at least three times.
