Cytotoxicity and ROS Evaluation of Cur

Cur (Mw = 368.38, purity ≥98%), sodium hydroxide, and trisodium citrate were purchased from Macklin Biochemical Co., Ltd. (Shanghai, China). The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was provided by Sigma-Aldrich Co. (St. Louis, MO). The dialysis bags were obtained from Sinopharm Chemical Reagent Co. Ltd. (Shanghai, China). Dulbecco’s modified Eagle medium (DMEM), penicillin–streptomycin, and fetal bovine serum (FBS) was purchased from Gibco BRL (Gaithersburg, MD). ROS detection kit was obtained from Bio Lebo Technology Co., Ltd. (Beijing, China). Hoechst 33342 was purchased from Beyotime Biotechnology (Nanjing, China). The water used in the experiments was deionized. Other reagents were of analytic grade. MDA-MB-231 and 4T1 were obtained from the American Type Culture Collection (ATCC) (Manassas, VA). Female Balb/c mice (4–5 weeks old) were obtained from the Laboratory Animal Center of Nantong University.

Carboxymethyl Chitosan Conjugation Protocol

CMCH was supplied by Zhejiang Yuhuan Biochemical Co. Ltd (China). Its carboxymethylation substitutive degree was 0.65 and the average molecular weight was 3.0 × 105 g/mol. In vitro F96 Release from Liposomes

F96 release from liposomes was investigated in vitro using dialysis membranes. Briefly, 4 mL of liposomes containing about 1 mg mL−1 F96 were placed in a dialysis bag (MWCO approximately 14 000, Sinopharm Chemical Reagent Co. Ltd, China). The dialysis bag was placed in a flask containing 100 mL release medium (PBS, pH 7.4, containing 0.2% Tween-80), and placed in a water bath shaking at 100 rpm at 37 °C. At predetermined time intervals, 1 mL of the release medium was withdrawn and replaced with an equal volume of fresh medium. The concentration of F96 in the medium was determined by high-performance liquid chromatographic (HPLC) and used to calculate the cumulative release percentage of F96.

In Vitro Drug Release Kinetics

HA/CNPs were collected and then suspended in 3 ml PBS containing 1% sodium dodecyl sulfate (SDS) (0.2 mol/l, pH 6.6) to adjust the drug concentration to 0.4 mg/ml. The final solution was put in a dialysis bag (Sinopharm Chemical Reagent Co., Ltd., Shanghai, China) (Mr2000). The dialysis bag was then placed in a beaker with 30 ml PBS and incubated in a water bath at 37°C, while shaking at 60 × g. A total of 1 ml of the solution was collected after 1, 2, 4, 8, 12, 18, 24, 36, 48, 60 and 72 h (isothermal medium was appended), and the OD values at 430 nm were measured. These values were drawn into a standard curve and used to calculate related concentrations and total release rates of the samples in two groups at each time point.

Dialysis bag (Mw = 8000 to 12 000 Da), dichloromethane (DCM), trifluoroacetic acid (TFA) and triethylamine (TEA) were obtained from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). Dimethyl sulfoxide (DMSO), 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) were all provided by Sigma-Aldrich (MO, USA). Poly(ethylene glycol) methyl ether (MeO-PEG-OH, MW = 2000 kDa) were ordered from TCI (PA, USA). Roswell Park Memorial Institute (RPMI) 1640 Medium, fetal bovine serum (FBS), penicillin-streptomycin solution, Dulbecco's phosphate-buffered saline (DPBS) and trypsin–EDTA (0.25%, w/v) solution were purchased from Invitrogen (NY, USA). All solvents used in this study were HPLC grade.
