Annexin V-fluoresceine isothiocyanate (FITC) Apoptosis Detection Kit (Beyotime) was exploited to detect the apoptotic rate of NCI-H929, MM1.S and RPMI-8226 cells. 2 × 105 cells seeded in 6-well plate were incubated for 24 h. Later, 10 μL Annexin V-FITC and propidine iodide (PI) solution were added. 15 min later, apoptotic cells were measured by a flow cytometer (Beckman Coulter, Fullerton, CA, USA) and analyzed using FACScan. The apoptotic rate was computed by the analysis of the proportion of cells at Annexin V +/PI ± .

The cells were inoculated into 6-well plates. The adherent cells were digested with trypsin. The suspended cells were collected and the concentration was adjusted to 3 × 106/ml. The cell apoptosis was then detected with Annexin V- Fluoresceine Isothiocyanate (FITC) apoptosis detection kit (Beyotime, China). Specifically, 5 μl Annexin V-FITC, and 5 μl propidium iodide were added, respectively; then, the cells were staining for 15 min, and the flow cytometry was applied to measure the apoptosis ratio within 1 h.
