Mycobacterium tuberculosis H37Ra Drug Susceptibility

Mycobacterium tuberculosis H37Ra was grown at 37°C in Middlebrook 7H9 liquid medium or on 7H11 agar plates supplemented with 10% (v/v) albumin–dextrose–catalase (ADC, Becton Dickinson, Sparks, MD, United States) plus 0.5% (v/v) glycerol, 0.25% (v/v) Tween 80. Plasmids p2NIL and pGOAL19 used in this study were obtained from Addgene (Cambridge, MA, United States). isoniazid (INH), rifampicin (RIF), streptomycin (SM), ethambutol, pyrazinamide (PZA), levofloxacin, amikacin, cycloserine, p-aminosalicylic acid (PAS), clofazimine (CFZ), tetracycline, linezolid, clarithromycin, and piperine were purchased from Sigma-Aldrich (St Louis, MO, United States). The drugs were dissolved in DMSO and further diluted to obtain the desired concentrations in culture media for drug susceptibility testing (see below).

Antimicrobial Susceptibility Testing Protocol

Triclosan, INH, ethambutol, rifampicin, clofazimine, ethionamide, p-aminosalicylic acid, D-cycloserine, tetracycline, erythromycin, gentamicin, ofloxacin, levofloxacin, acriflavine, thioridazine hydrochloride, reserpine, o-vanadate, CCCP, verapamil, and cetyltrimethyl ammonium chloride (CTAB) were obtained from SIGMA. Streptomycin, amikacin, and kanamycin were obtained from Apollo Chemicals Ltd. Chloramphenicol was obtained from Boehringer Mannheim, hygromycin B from Roche, safranin from Panreac, and ethidium bromide from Amresco. Stocks were prepared according to the manufacturer’s instructions, and aliquots kept at −20 °C were thawed just before use. For the MIC assays, drugs were diluted to the working concentration in Middlebrook 7H9 supplemented with 0.2% glycerol and 10% ADC.

Quantitative Analysis of Drug Compounds

Acetaminophen, clofazimine, D-cycloserine, ethyl acetate, isoniazid, levofloxacin, linezolid, meropenem, moxifloxacin, p-aminosalicylic acid, pyrazinamide, rifampicin, sodium chloride and trimethoprim were purchased from Sigma-Aldrich, Australia. Abacavir, azithromycin, emtricitabine, ethambutol, ethionamide, fluconazole, lamivudine, nevirapine, sulfamethoxazole and zidovudine were purchased from Sapphire Bioscience, Australia. All chemicals were of !98% purity. Ultrapure water (resistivity at 25 C = 18.2 MXÁcm) was obtained using a Milli-Q V R Integral Water Purification System (Merck Millipore, Australia).
The samples were measured on a UV spectrophotometer, NanoPhotometer NP80 V R (Implen, Germany), using disposable cuvettes (70-850 lL, 8.5 mm, Brand V R UV cuvettes). The UV spectrophotometer was set on wavescan mode with a detection range of 200-900 nm, pathlength of 10 mm, baseline correction and smoothing filter switched off.
