Poly(DL-lactide-co-glycolide) (PLGA, 50:50, inherent viscosity 0.59 dL/g, MW 53.4 kDa) and poly(L-lactide) (PLA, inherent viscosity 1.16 dL/g, MW 158.0 kDa) were purchased from Durect Corporation, Birmingham, UK. Ketorolac (MW 255.27 g/mol) in the form of Ketorolac tromethamine (water solubility 200 mg/mL, log P 2.1 [22 ]) was obtained from Cayman Chemical Company, Ann Arbor, MI, USA. Ltd., Singapore), acetone (Honeywell Burdick & Jackson, Morris Plains, NJ, USA), dichloromethane (DCM, Honeywell Burdick & Jackson, Morris Plains, NJ, USA), methanol (high-performance liquid chromatography (HPLC) grade, Honeywell Burdick & Jackson, Muskegon, MI, USA), and sterile water for irrigation (SWI, General Hospital Products. Public Co., Ltd., Pathum Thani, Thailand) were used as received.

Multimodal Analgesic Efficacy Assessment

Morphine (Hikma), ketorolac (Cayman Chemical), and gabapentin (Acros Organics) were administered in a volume of 1 μl/g body weight. Morphine (1 mg/kg, dissolved in sterile saline) was given as a subcutaneous injection while ketorolac (10 mg/kg, dissolved in sterile saline) and gabapentin (100 mg/kg, dissolved in sterile saline) were given as intraperitoneal injections. Subcutaneous Osmotic Pump Implantation

Two days following vasculature imaging, osmotic pumps (Alzet, Cupertino, CA cat.no. 2002) were implanted subcutaneously according to the manufacturer's instructions under isoflurane anesthetization (1%–3%). Pumps were filled with either saline or ketorolac (Cayman Chemical Company, Ann Arbor; MI cat.no. 70690) dissolved in saline. Pumping rate was 0.5 µl/hour. The ketorolac concentration was adjusted to the animal weight to achieve 0.64, 1.28, and 6.4 mg/kg/hour, and was 0.0255, 0.0512, and 0.256 mg/µl, respectively, for a 20‐g mouse.
