Immunohistochemical Analysis of Protein Expression

Samples were fixed with 4% paraformaldehyde, dehydrated through a graded series of ethanol, paraffin-embedded, and sliced into 5-mm sections. Protein Expression Profiling by Western Blot

Total protein lysates extracted from samples were separated with 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and transferred to a polyvinylidene fluoride (PVDF) membrane. The membrane was blocked with 3% bovine serum albumin BSA, followed by incubation with antibodies. The membrane was then incubated with secondary horseradish peroxidase (HRP)-conjugated goat anti-rabbit or anti-mouse antibody (Jackson ImmunoResearch Laboratories Inc., West Grove, PA, UAS), and visualized using Immobilon™ Western Chemiluminescent HRP substrate (Millipore, Burlington, MA, USA). Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was used as the loading control.
