Human T cells were purified from peripheral blood mononuclear cells of healthy donors with an EasySep Human T-cell Enrichment kit (STEMCELL Technologies Inc., Vancouver, BC). They were then activated by human T-Activator DanyBeads (Life Technologies, Grand Island, NY) and expanded in complete RPMI-1640 medium in 96-well plates (105 cells/ml) in the presence of recombinant human IL-2 (20 ng/ml) (PeproTech, Rocky Hill, NJ) for 4 days. After washing with RPMI-1640 medium, T cells were co-cultured at a 3:1 or 2:1 ratio with PLX4032- or DMSO-pretreated cancer cells (2×104 cells/well) in a 96-well plate for 24 h. Alternatively, activated T cells were pre-incubated with an anti-human TRAIL neutralizing antibody for 1 h at 4°C before being utilized for co-culture with the tested cancer cells.
