4 hydroxynonenal hne

Manufactured by Cayman Chemical
Sourced in Germany

4-hydroxynonenal (HNE) is a key lipid peroxidation product that serves as a marker of oxidative stress. It is a highly reactive α,β-unsaturated aldehyde that can form adducts with proteins, DNA, and other biomolecules. HNE is widely used in biochemical research to study the effects of oxidative stress and its role in various pathological conditions.

Human glioblastoma cell lines LN229, U87MG, and T98G and glioblastoma stem-like cells X01 and T84 were cultured as previously described [12 (link)]. TMZ, N-acetylcysteine (NAC), and bafilomycin were obtained from Sigma (Munich, Germany). 4-Hydroxynonenal (HNE) was from Cayman Chemical (Hamburg, Germany). Antibodies against HNE were obtained from Abcam (England, UK); ALDH1A3 (mouse) and LC3 from Novus Biologicals (Abingdon, UK); ADLH1A3 (rabbit) and p62 from Thermo Fisher Scientific (Munich, Germany); and phospho-H2A.X (Ser139), vinculin, and HRP-conjugated secondary antibody from Cell Signaling Technology (Frankfurt, Germany).

Preparation and Labeling of α-Synuclein Oligomers

Check if the same lab product or an alternative is used in the 5 most similar protocols
Recombinant α-SYN (140 μm) was produced as described previously (Näsström et al., 2011 (link)). Monomeric α-SYN was incubated with 4-hydroxy-nonenal (HNE) (Cayman Chemicals) in a HNE: α-SYN ratio of 30:1 at 37°C for 72 h. SEC-HPLC was performed on a Superose 6 PC 3.2/30 column (GE Healthcare) and revealed a near complete conversion from monomers to oligomers (Fig. 1-1B). As an eluent, 20 mm Tris and 0.15 m NaCl, pH 7.4, was used at a flow rate of 50 μl/min. The α-SYN oligomers were labeled with the Cy3AM Antibody Labeling Kit (GE Healthcare, PA33000), the Atto 488 protein labeling kit (Sigma-Aldrich) or pHrodo (Invitrogen, P36600). The pHrodo dye reacts with amine groups on the cell surface and is nonfluorescent at neutral pH, but emits red light at an increasing intensity as the pH is lowered. For all labeling, the protocols provided with the dye kits were followed and unbound excess Cy3, Atto 488, or pHrodo were removed by filtration in a Zeba spin desalting column (Thermo Scientific) according to the manufacturer's instructions.

Carbonyl cyanide 4-(trifluoromethoxy)phenylhydrazone (FCCP) and calyculin A were from Santa Cruz Biotechnology. Anti-vimentin antibody (sc-6260) and its Alexa-488 and agarose conjugates were from Santa Cruz Biotechnology, anti-phospho-Ser56 and anti-phospho-Ser72 vimentin antibodies were from MBL, and anti-actin was from Sigma. About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
