Calcium Imaging Assay Protocol

Cal-520 in acetoxymethyl (AM) ester form (Abcam, Cambridge, MA) was utilized as the primary Ca2+ indicator, which was reconstituted in dimethyl sulfoxide (DMSO) at a concentration of 1 mM and stored at −20 °C. DMSO, probenecid, and sodium butyrate were obtained from Millipore-Sigma (St. Louis, MO). Rat tail type I collagen, polystyrene tissue culture plates (6-well, 12-well, and 24-well), 12-well Transwell inserts, and HEPES were purchased from Corning (Corning, NY). DAPT was obtained from Xcess Biosciences (Chicago, IL). Recombinant forms of bone morphogenetic protein 2 (BMP-2) and bone morphogenetic protein 9 (BMP-9) were obtained from R&D Systems (Minneapolis, MN). Deionized (DI) water (≥17.8 MΩ·cm), purified through a Barnstead NANOpure Diamond (Thermo Scientific) filtration system, was used for all reagent preparations.

Hyaluronic Acid Conjugate Synthesis

Desalting PD10 columns were purchased from GE Health-care Bio-Sciences (Pittsburgh, PA, USA). D2O [99.9% D] and DMSO-D6 [99.8% D] were purchased from EMD Millipore. All water was obtained from a Barnstead NANOpure Diamond or Barnstead GenPure system (Thermo Fisher Scientific) producing 18.2 MX water.

Chemically Modified Hyaluronic Acid Nanoparticles

HA was purchased from LifeCore Biomedical (Chaska, MN). 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) N-hydroxysuccinimide (NHS), N,N-dimethylformamide (DMF), dimethyl sulfoxide (DMSO), methanol, dialysis membranes (3,500 Da MWCO and 6,000-8,000 Da MWCO), gelatin powder N.F. grade, and TRIS-HCl were purchased from Fisher Scientific. 5β-cholanic acid, 1-pyrenebutyric acid, sodium azide, dried erythrocytes, 1,3-diaminopropane, and ICG were purchased from Sigma Aldrich (St. Louis, MO). Cy7.5-amine was purchased from Lumiprobe Corporation (Hallandale Beach, FL). All the reagents were used without further purification unless otherwise specified. MEM, fetal bovine serum, phosphate buffered saline, and penicillin/streptomycin were obtained from Cell and Viral Vector lab at Wake Forest Health Sciences or from the General Supply at the University of Nebraska Medical Center. All water was obtained from a Barnstead NANOpure Diamond or Barnstead GenPure system (Thermo Scientific; Waltham, MA) producing 18.2 MΩ water. Human breast cancer cell line MDA-MB 231 and mouse breast cancer 4T1 were obtained from ATCC (Manassas, VA).

Delamanid Formulation Development and Characterization

was purchased from Gojira Fine Chemicals, LLC (Bedford Heights, OH).
Hydroxypropyl methylcellulose (HPMC; Pharmacoat 603), hydroxypropyl
methylcellulose phthalate (HPMCP; HP-50), and hydroxypropyl methylcellulose
acetate succinate (HPMCAS; AQOAT AS-LF and AS-HF) were generously
provided by Shin-Etsu Chemical Co. (Tokyo, Japan). Dichloromethane
(DCM; HPLC Plus grade) and Tween 80 were purchased from Sigma-Aldrich
(St. Louis, MO). Polycaprolactone diol (PCL; MW 1250 Da) was purchased
from Polysciences, Inc. (Warrington, PA). l-α-Lecithin,
anisole (99%), and methyl tert-butyl ether (MTBE;
99.9%) were purchased from Acros Organics (Geel, Belgium). Tetrahydrofuran
(THF; HPLC grade), acetonitrile (HPLC grade), methanol (HPLC grade),
chloroform (HPLC grade), ethyl acetate (ACS grade, 99.9%), sodium
hydroxide, and trifluoroacetic acid were purchased from Fisher Scientific
(Hampton, NH). N-butyl acetate (>99%) was purchased from Alfa Aesar
(Ward Hill, MA). HEPES (4-[2-Hydroxyethyl]-1-piperazine ethanesulfonic
acid) was purchased from Boehringer Mannheim (Mannheim, Germany).
Deionized (DI) water (18.2 MΩ cm) was prepared by a Barnstead
Nanopure Diamond ultrapure water system (Thermo Scientific, Waltham,
MA).

Quantification of Furosemide and Hydrochlorothiazide

Standard solutions of furosemide and hydrochlorothiazide (HCT; purities ≥98% and ≥99%, respectively) were obtained from Sigma-Aldrich (St. Louis, MO, USA). Methanol (purity LC–MS) was obtained from JT Baker® (Avantor Performance Materials, Inc., Center Valley, PA, USA). Formic acid (purity approximately 98%) was obtained from Sigma-Aldrich. The water used, HPLC-grade, was obtained from distilled water for purification, via the purification system Barnstead Nanopure Diamond (Thermo Fisher Scientific, Palo Alto, CA, USA). Stock solutions of furosemide and HCT (used as internal standard) were obtained by dissolving 15.0 mg of standard solids, accurately weighed, in 100 mL of methanol, obtaining solutions of 150 mg L−1 (150 ppm) concentration, which were stored at −20 °C. Working solutions used for the analysis in LC–MS/MS or for the creation of urine samples fortified were obtained starting from stock solutions by dilution with water or methanol, using a dilution factor less than 10 in each step of dilution.

Nanopure Water Purification Protocol
