Chidamide and Chemotherapeutics Combination

Check if the same lab product or an alternative is used in the 5 most similar protocols
Chidamide (CS055, purity >95%) was supplied by Chipscreen Bioscience Ltd. (Shenzhen, China) and dissolved in dimethyl sulfoxide (DMSO) (Invitrogen, USA) to obtain a stock solution of 50 mM. Idarubicin (IDA), daunorubicin (DNR), and cytarabine (Ara-C) were purchased from Sigma-Aldrich (St Louis, MO) and dissolved in phosphate-buffered saline (PBS) at the concentrations of 10, 100, and 100 mM, respectively. All the above stock solutions were stored at −20 °C. It was diluted to the required concentrations in subsequent experiments with culture medium.

Pharmacological Inhibitors in FLT3-ITD Signaling

Check if the same lab product or an alternative is used in the 5 most similar protocols
The pan-Pim kinase inhibitor AZD1208, from Tocris Bioscience (Minneapolis, MN, USA), was used at 1 μM based on phase I clinical trial data [10 (link)] and on inhibition of BAD phosphorylation at serine 112 as a pharmacodynamic endpoint [37 (link)]. The topoisomerase 2 inhibitors daunorubicin (DNR), mitoxantrone (MXR) and etoposide (VP-16) (Sigma-Aldrich) were dissolved in dimethylsulfoxide (DMSO) (less than 0.05 percent) and used at their approximate IC50 concentrations of 10, 20 and 30 nM, respectively, in FLT3-ITD cell lines [13 (link)]. The protein translation inhibitor cycloheximide (CHX) was from Sigma-Aldrich and the proteasome inhibitor carbobenzoxy-L-leucyl-L-leucyl-L-leucinal (MG-132) from Calbiochem (San Diego, CA, USA). The c-Myc inhibitor 5-[(4-ethylphenyl)methylene]-2-thioxo-4-thiazolidinone (10058-F4), which inhibits c-Myc-Max binding and thereby prevents transcriptional activation of c-Myc targets, was also from Sigma-Aldrich.

Lentiviral sh-coilin Knockdown in Reh and RS4;11 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Reh and RS4;11 cells were purchased from the cell bank at Peking Union Medical University. The Reh cell line was detected for unique STR profiles in China and confirmed with the correct genetic background. Cells were expanded and cultured in Roswell Park Memorial Institute (RPMI) with l-glutamine (Hyclone, Beijing, China) supplemented with 10% fetal bovine serum (FBS) (Invitrogen, Carlsbad, CA). For lentivirus infection experiments, cells were seeded in 6- or 12-well plates. The lentivirus carrying a sh-coilin cassette (4 × 108 TU/ml) was purchased from Shanghai Genechem Co., Ltd, China. The targeting RNAi sequence of sh-coilin and a control scramble were as GAGAGAACCTGGGAAATTT and TTCTCCGAACGTGTCACGT respectively.
Daunorubicin (DNR, Sigma-Aldrich, USA) was used at 0.01, 0.03 and 0.05 μg/ml for 24 h. Arabinocytidine, Vincristine and Adriamycin were purchased from Dalian Meilun Biotechnology Co., Ltd, China.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
