Isolation and Culture of A. alternata

The wild type (WT) A. alternata strain JT-03 was isolated from infected pear fruit and cultured on potato dextrose agar (PDA) at 28 °C for 5 d. The pCAMBIA1300-HPH and pC-NEO-NGFP vectors were provided by the Chinese Academy of Sciences. The pear wax used in the experiments was extracted from pears as described by Yin et al. [42 (link)] and then prepared as a 0.1% wax solution. KN93 and Gelbond PAG film were purchased from TopScience (Shanghai, China) and Univ-bio (Shanghai, China). The pear (Pyrus bretchneideri ‘Zaosu’) was harvested from a Tiaoshan Farm in Jingtai County, Gansu Province, China.
