Cell Cycle Analysis of Prostate Cancer Cell Lines

RWPE-2, PC3, DU145, and MDA-PCa-2b cells were serum-starved overnight and transfected with NC or miR-214 mimic for 48 h and then harvested by trypsinization. Cells were washed in ice-cold phosphate-buffered saline (PBS) and fixed in ice-cold 70% ethanol overnight at −20 °C. Cell pellets were stained with FxCycl PI/RNase staining solution (Life Technologies Corporation) for 15 min at room temperature in the dark. Cell Cycle Analysis by Flow Cytometry

Cells were harvested and fixed with 70% ethanol at 4 °C overnight; then cells were washed and stained with 0.5 mL of FxCycle PI/RNase Staining Solution for 30 min at room temperature in the dark. Finally, the cell cycle was measured using a flow cytometer (Guava EasyCyte HT, EMD Millipore, Billerica, MA, USA), and the data were analyzed using Flow Jo software (FLOWJO, Ashland OR, USA).
