Anthropometry was evaluated and laboratory tests were performed in all the participants according to established protocols at OPBG. Participants were asked to refrain from intensive physical activity in the 3 days prior to any study. Weight and height were measured using standard procedure. Up to 3 blood pressure measurements were taken by a physician after a 5‐min rest and the mean of 3 measurements was recorded.¹³Blood samples were drawn after 8–12 h fast. A standard OGTT (1.75 g/kg body weight up to a maximum of 75 g) was performed with flavoured glucose (Glucosio Sclavo Diagnostics, 75 g/150 ml). One anti‐cubital i.v. catheter was inserted for blood sampling and was maintained patent by a normal saline drip during the test. Glucose level was measured by glucose oxidase technique (Cobas Integra, Roche) and insulin by a chemiluminescent immunoassay method (ADVIA Centaur analyser; Bayer Diagnostics). HbA1c was measured using an automated high performance liquid chromatography VARIANT II TURBO Haemoglobin Testing System (Bio Rad, Hercules, CA).

Platelet-depleted serum concentration of 5-HT was measured in duplicate using Serotonin FAST Kit (catalog no. EIA5061; DRG instrument GmbH, Marburg, Germany). Its characteristics are as follows: detection range=15–2,500 ng/mL; sensitivity=5 ng/mL.
An aliquot of aspirated platelet rich serum was then used to assess platelet count via Advia Centaur Analyser (Bayer Ltd., Berks, UK). Additional duplicate samples were used for 5-HT analysis of platelet-rich serum, while platelet-depleted serum 5-HT concentrations were measured following centrifugation.
