Intracellular ROS Measurement Protocol

The level of intracellular ROS generation was assessed using the ELISA kit (Shanghai Yanjing Biological Research Technology Co, LTD, China) according to manufacturer's instruction. Furthermore, fluorescence microscopy was used to confirm the results of the microplate ROS assay. Briefly, after the indicated treatments, cells were washed twice with phosphate-buffered saline and then incubated with serum-free DMEM containing 10μmol/l dichlorofluorescein diacetate (DCFH-DA) at 37˚C for 30 min. After that, cells were washed with PBS three times in order to eliminate the residual DCFH-DA. Cells from each group were analyzed by measuring the excitation and emission spectrum at 488 and 525 nm, respectively. The fluorescence was visualized using a fluorescence microscope coupled with an image analysis system Olympus IX71 system biological microscope.
