HPLC Analysis of Antibiotic Standards

OTC (CAS No. 79-57-2, purity ≥ 97.0%) and TC (CAS No. 60-54-8, purity ≥ 98.0%) standards were purchased from Sigma-Aldrich LLC (St. Louis, MO, USA). A DOX (CAS No. 564-25, purity ≥ 98.0%) standard was acquired from Merck Drugs & Biotechnology Co., Inc. (Fairfield, OH, USA). CIP (CAS No. 85721-33-1, purity ≥ 98.0%) and ENR (CAS No. 93106-60-6, purity ≥ 98.0%) standards were obtained from Macklin Inc. (Shanghai, China).
LC-grade methanol and acetonitrile were provided by Tedia Company Inc. (Fairfield, OH, USA). Sodium hydroxide, citric acid, magnesium chloride, malonic acid and other reagents were of analytical grade and were supplied by Sinopharm Chemical Reagent Co. Ltd. (Shanghai, China). Sodium dodecyl sulfate was obtained from Solarbio Life Science Co., Ltd. (Beijing, China). Ultrapure water (18.2 MΩ* cm, 25 °C), which met the standard of ‘water for analytical laboratory use-specification and test methods’ [12 ], was prepared with a Nanopure water purifier produced by Thermo Fisher Scientific Inc. (Waltham, MA, USA). All solutions used in the LC system were filtered through a 0.45 μm pore size polytetrafluoroethylene microporous membrane filter and then degassed.

Glucuronidation of Fluorogenic Probe

The fluorogenic probe substrate N-butyl-4-(4-hydroxyphenyl)-1,8-naphthalimide (NHPN) and its O-glucuronide (NHPNG) were synthesized and purified (purity≥98%) according to the previously reported scheme [13 (link)]. Nilotinib and chrysin were from Dalian Meilun Biotech Co., Ltd. (Meilun, Dalian, China). The tissue preparations, including pooled human liver microsomes (HLM), pooled human intestine microsomes (HIM), pooled human kidney microsomes (HKM), and pooled human lung microsomes (HLuM) were purchased from Celsis Inc. (Baltimore, MD, USA). Recombinant human UGT1A1 was from BD Biosciences (Woburn, MA, USA). Stably transfected HeLa cells (named HeLa-UGT1A1 cells) were constructed as previously reported [[15] (link), [16] (link), [17] ]. The uridine diphosphate-glucuronic acid (UDPGA), polyethylene glycol hexadecyl ether (Brij 58) and anti-GAPDH antibody (G8795) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Anti-UGT1A1 antibody (ab194697) was obtained from Abcam (Cambridge, MA). The reagents for the SimpleWestern blotting system were purchased from ProteinSimple (San Jose, CA). Ultrapure water purified by Milli-Q® Integral Water Purification System (Millipore, USA) was used throughout, while LC grade methanol, acetonitrile, and formic acid were ordered from Tedia (Fairfield, USA).

Trace-level Pesticide Extraction

LC-grade methanol and acetonitrile were purchased from TEDIA. The analytical standards (ethoprophos, coumaphos, profenofos, diazinon, ethion, parathion, phosalone, quinalphos, dicrotophos, purity >95%) were purchased from Sigma Aldrich. Azinphos methyl, tribuphos (purity >95%) were from Dr. Ehrenstorfer Gmbh. Other details are given in Table 1. The analytical-reagent grade anhydrous magnesium sulfate (MgSO4), sodiumchloride (NaCl) was from Kermel. The powder of C18 and primary secondary amine (PSA, 50 μm) were purchased from Agela Technologies. The guarantee dreagent-grade formic acid was purchased from ROE. Ultrapure water was obtained from a Milli Q purification system (Millipore, Molsheim). Individual pesticide stock standard solutions (1000 mg L -1 ) were prepared in methanol and stored at -18°C. A multistandard mixture containing 10 mg L -1 of each pesticide was prepared in methanol and stored at 4°C for less than 3 months. Mixed multi-standard working solutions were prepared fresh to avoid degradation of pesticides. The concentrations of the multi-standard working solutions were 10, 25, 50, 100 and 250 μg kg -1 .
