Evaluating Blood-Brain Barrier Integrity

BBB integrity was analysed by evaluating Evans blue extravasation. For this, 1-year-old Cort+/+ and Cort−/− mice were injected intraperitoneally with LPS from E.coli 055:B5 (6 mg/kg, Sigma) inducing mild neuroinflammation [20 (link)]. Immediately after LPS injection, 1 nmol of CST-29 (resuspended in PBS, Bachem) was intraperitoneally injected. 5 h after LPS and LPS + CST injection, Evans Blue (EB, 2% in saline, 4 ml/kg) was injected through the tail vein. Control animals were injected with vehicle (PBS). After 1 h, mice were sacrificed by intraperitoneal injection of pentobarbital and intracardially perfused with PBS. EB quantification was performed as indicated in Additional file 1. Data were represented as μg of dye per mg of brain tissue.
