Quantification of Drug Transporter Expression

Mouse monoclonal antibodies against P-gp (clone C219), MRP2 (clone M2III-6) and MRP3 (ABCC3) (clone M3II-9) were from Alexis Biochemicals (Lausen, Switzerland), whereas rabbit antibody against the p38 mitogen-activated protein kinase (MAPK) was provided by Santa Cruz Biotechnology (Dallas, TX, USA). Mouse monoclonal antibody against MRP4 (ABCC4) and rabbit polyclonal antibody against MRP5 (ABCC5) were purchased from Abcam (Paris, France). All other chemicals and reagents were commercial products of the highest purity available.

TCDD and GNF-351 Cytotoxicity Assay

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD, 50 μg/mL in DMSO, >99.9%) was purchased from Cambridge Isotope Laboratories (Tewksbury, MA) and GNF-351 was purchased from Santa Cruz Biotechnology (Dallas, TX). Trichloroacetic acid was purchased from Merck (Darmstadt, Germany). Resorufin ethyl ether, 3,3′-methylene-bis(4-hydroxycoumarin), sulforhodamine B and DMEM/F12 powder were purchased from Sigma (St Louis, MO). 100x L-glutamine and penicillin-streptomycin were purchased form Gibco (Dublin, Ireland) while fetal bovine serum was purchased from Cytiva Life Sciences (Marlborough, MA). HepG2, Hepa1c1c7 and H4IIE cell lines were purchased from ATCC (Manassas, VA).

Assessing AhR Signaling in 4HNE Cytotoxicity

To determine the role of AhR signaling in maintaining cell viability in light of 4HNE-mediated cytotoxicity, cells were plated in a 96-well format with a density of 150,000 cells per plate. Cells were then treated with desired concentrations of 2,3,7,8-tetrachlorodibenzo-p-dioxin (Cambridge Isotope Laboratories), 2-(2-aminophenyl) (Sigma-Aldrich), palmitoleic acid (Cayman Chemical Company) or control DMSO (Sigma-Aldrich). In determining viability dependence on AhR, cells were co-treated with AhR ligands and desired concentrations of α-naphthoflavone (Sigma-Aldrich). Molecule treatments lasted for 48 hours and were and co-treatment with 4-hydroxynonenal (Cayman Chemical Company) for 24 hours. At which point, cell viability was assessed using the CellTiter-Glo luminescent cell viability kit (Promega. Inc) and a Fluoroskan Ascent FL luminometer. Cell viability was also accessed using Calcein-AM kit (Life. Inc.) and a Spectramax fluorescence plate reader.

Preparation and Characterization of Polychlorinated Biphenyl Standards

2,2′-Dichlorobiphenyl (PCB 4), 2,4′-dichlorobiphenyl (PCB 8), 3,3′-dichlorobiphenyl (PCB 11), 2,2′,5-trichlorobiphenyl (PCB 18), 2,4′,5 -trichlorobiphenyl (PCB 31) and 2,2′,5-trichlorobiphenyl-4′-ol (4′-OH-PCB 18) were purchased from AccuStandard (New Haven, USA). 2,4,4′-Trichlorobiphenyl (PCB 28) was from Dr. Ehrenstorfer (LGC Standards, Teddington, UK). 2,4′-dichlorobiphenyl-4-ol (4-OH-PCB 8), 3,3′-dichlorobiphenyl-4-ol (4-OH-PCB 11) and 3,3′-dichlorobiphenyl-5-ol (5-OH-PCB 11) were synthesized as described in Supplementary Experimental Procedures. The purities of all LC-PCB congeners and trace impurities are reported in Supplementary Tables 1 and 2. 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD), 99% purity, was from Cambridge Isotope Laboratories (Andover, USA). ICI 182,780 was from Tocris Bioscience (Bristol, UK). Mifepristone was purchased from (Cayman Chemical). Cyproterone acetate (CPTA) was from Santa Cruz Biotechnology (Santa Cruz, USA). 17β-Estradiol (E2), 5α-dihydrotestosterone (DHT), triiodothyronine (T3), rosiglitazone (RGS), phenobarbital (PB) and rifampicin (RIF) were from Sigma-Aldrich (Prague, Czech Republic). All other chemicals not specified above were of the highest purity available; they were purchased from Sigma-Aldrich, if not indicated otherwise.
