Fitc conjugated anti cd4

Manufactured by Beckman Coulter
Sourced in France

FITC-conjugated anti-CD4 is a monoclonal antibody conjugated with the fluorescent dye FITC (Fluorescein Isothiocyanate). It is designed to detect and label the CD4 cell surface antigen, which is primarily expressed on helper T cells.

Multiparametric Flow Cytometry Analysis

FITC-conjugated anti-CD4, PE-conjugated anti-IFN-γ, anti-IL-17A, and anti-IL-21 mAbs were purchased from Beckman Coulter (Marseilles, France), eBioscience (San Diego, CA, USA), and BD Biosciences (San Jose, CA, USA), respectively. For cytokine detection, 500 μL fresh heparinized peripheral blood was incubated in an equal volume RPMI 1640 medium, and stained with anti-CD4, and then activated with 50 ng/mL phorbol 12-myristate 13-acetate (PMA), 1 μg/mL ionomycin (Sigma, St Louis, MO, USA) and 0.7 µL GolgiStop (BD Biosciences) simultaneously for 4 h. After fixation and permeabilization, cells were stained with the above-mentioned specific antibodies. At least 1 × 105 events in the lymphogate were collected by flow cytometry (Beckman Coulter Cytomics FC500) for analysis.

Multicolor Flow Cytometry of Lymphocytes

A three-or four-color flow cytometric analysis was performed for at least 20,000 lymphocytes by using an EPICS XL (Beckman Coulter) as described previously (Honda et al. 2000) . Phycoerythrin (PE)-conjugated anti-CD3 antibody (BD Biosciences, Mountain View, CA, USA), fluorescein isothiocyanate (FITC)-conjugated anti-CD19 antibody (Beckman Coulter, Miami, FL, USA) and phycoerythrin-cyanin 5.1 (PC5)-conjugated anti-CD56 antibody (Beckman Coulter) were used to determine the proportions of T-cells, B-cells and natural killer (NK) cells, respectively, among gated lymphocytes. FITC-conjugated anti-CD4, PE-conjugated anti-CD3 and PC5-conjugated anti-CD8 antibodies (Beckman Coulter) were used to determine helper and cytotoxic T-cells.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
