Culturing SCLC and NSCLC Cell Lines

Human SCLC cell lines (DMS53, NCI-H209, NCI-H69, SHP77, NCI-H889, NCI-H345, and NCI-H82) and human NSCLC cell lines (A549, NCI-H23, NCI-H460, NCI-H1155, NCI-H358, NCI-H727, NCI-H1770, and NCI-H125) were maintained in RPMI 1640 (Invitrogen, Carlsbad, CA) supplemented with 10% fetal bovine serum, and 1% penicillin streptomycin, as previously described (Hong et al. 2013 (link)). All SCLC and the two NSCLC lines, NCI-H1155 and NCI-H727, were previously characterized as NE tumors (Carney et al. 1985 (link); Gazdar and Minna 1996 (link); Linnoila 1996 (link)). Human primary diploid fibroblasts IMR90 were maintained in MEM (Invitrogen, Carlsbad, CA) supplemented with 10% bovine growth serum, 1% sodium pyruvate, 1% MEM non-essential amino acids, and 1% penicillin streptomycin. Vandetanib was purchased from Invitrogen and LC Laboratories (Woburn, MA). Cabozantinib was purchased from Selleckchem (Huston, TX).

Cell Line Maintenance and Reagent Protocols

The human MTC lines, TT and MZ-CRC-1, were maintained as previously described [19 (link)20 (link)21 (link)]. Briefly, TT was maintained in RPMI 1640 (Invitrogen, Carlsbad, CA, USA) supplemented with 16% fetal bovine serum (FBS), 100 units of penicillin and 100 µg of streptomycin per mL. MZ-CRC-1 was maintained in high-glucose DMEM (Invitrogen) supplemented with 10% FBS in culture dishes coated with rat collagen (Sigma, St. Louis, MO, USA). All experiments were performed using cells within 10 passages from the point of acquisition. MKT-077, tetra-methyl-rhodamine ethyl ester perchlorate (TMRE), and N-acetyl-cysteine (NAC) were purchased from Sigma. 5-(and-6)-carboxy-2',7'-dichlorodihydrofluorescein diacetate (carboxy-H2DCFDA) and vandetanib were purchased from Invitrogen and LC Laboratories (Woburn, MA, USA), respectively.
