Characterization of P-glycoprotein modulators

The lipids 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC), 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoethanolamine (POPE), and 1-palmitoyl-2-oleoyl-sn-glycero-3-phospho-L-serine (POPS) were acquired from Avanti Polar Lipids, Inc. (Alabaster, AL, USA). [125I] Iodoarylazidoprazosin (IAAP) 1100 Ci/mmol) was purchased from PerkinElmer Life Sciences (Wellesley, MA, USA), and Tariquidar was purchased from AdooQ BioScience (Irvine, CA, USA). The fluorescent NBD amphiphiles were synthesized and purified, as previously reported by us [41 (link),42 (link)], and the products were stored at −20 °C in the dry state or dissolved in DMSO. The concentration of the NBD–amphiphiles was calculated from their absorption at the maxima (≅466 nm, ε = 2.1 × 104) [41 (link),42 (link)]. Reagents and solvents used were analytical grade or with higher purity, and water was distilled and further deionized with a final resistance ≥ 18 MΩ.
The structures and common molecular descriptors of the P–gp modulators characterized experimentally in this work are shown in Table 1.

Materials Propranolol hydrochloride, dexamethasone, and bis-p-nitrophenyl phosphate (BNPP) were obtained from Nacalai Tesque, Inc. (Kyoto, Japan). Metoprolol tartrate, atenolol, and acyclovir were obtained from Tokyo Chemical Industry Co., Ltd. (Tokyo, Japan). Nadolol and rhodamine 123 were obtained from Sigma-Aldrich Co. (St. Louis, MO, U.S.A.). Tariquidar was obtained from AdooQ BioScience (Irvine, CA, U.S.A.). Temocapril hydrochloride and temocaprilat were obtained from Toronto Research Chemicals, Inc. (North York, ON, Canada). All other chemicals were of the highest available purity.
