Quantitative Analysis of T-2 and HT-2 Toxins

Solvent purity LC-MS grade as methanol (MeOH), acetonitrile (ACN), 2-propanol (IPA), acetone (ACE), dimethylsulfoxide (DMSO) and standards of mycotoxins T-2 toxin and HT-2 toxin were purchased from Sigma-Aldrich (Prague, Czech Republic). The stock solution standards of mycotoxins (100 µg/mL in acetonitrile) were prepared in 10% methanol/water (v/v) at a concentration of 1 µg/mL and stored at 4 °C in the refrigerator before use. Sodium chloride was purchased from Lachner (Neratovice, Czech Republic). Additives for LC-MS (purity LC-MS) as ammonium acetate, formic acid, acetic acid and citric acid (purity ≥ 99.5%) were purchased from Sigma-Aldrich (Prague, Czech Republic). Check-Sample-Survey, milk thistle powder obtained from Central Institute for Supervising and Testing in Agriculture (ÚKZÚZ Brno, Czech Republic). Ultrapure water was produced by Aqua Osmotic 06 (Tišnov, Czech Republic). Immunoaffinity columns R-Biopharm EASI-EXTRACT® T-2 & HT-2 were obtained from Jemo Trading (Bratislava, Slovak Republic).

Curcumin-loaded Polymeric Nanoparticles

Pesticide Analysis in Citrus Fruits

The pesticide standards were purchased from Restek (Bellefonte, Pennsylvania, USA) and Lab Instruments (Castellana Grotte, Italy). HPLC-MS-grade acetonitrile, methanol, formic acid, and acetic acid were obtained from Lachner (Neratovice, Czech Republic). Sodium chloride, magnesium sulfate anhydrous, disodium hydrogen citrate sesquihydrate (C6H8Na2O8), and trisodium citrate dihydrate (C6H5O7Na3 × 2H2O) were obtained from Lachner (Czech Republic). The graphitized carbon black (GCB), C18 sorbent, and primary and secondary amines (PSA) were supplied from Sigma-Aldrich (St Louis, Missouri, USA).
The selected citrus fruits were purchased from local markets in Belgrade, Serbia, in February 2022, and a total of 76 samples were analyzed (28 oranges, 26 lemons, 17 tangerines, and 5 grapefruits). Each fruit sample was analyzed as the whole fruit following European Union Guidelines Regulation, (EC) No. 396/2005 Annex [20 ]. During the survey, fruits were stored in fridges in labeled plastic bags at 4 °C until analysis.

Bacterial Culture Media Preparation

All compounds were commercial products: hydrochloric acid, sodium chloride, and sodium hydroxide (Lach-Ner, Neratovice, Czech Republic), sodium and potassium phosphates, (Penta, Praha, Czech Republic), tryptone (Oxoid, Basingstoke, UK), yeast extract, kanamycin, and tetramethoxysilane (TMOS) (Sigma-Aldrich, St. Louis, MO, USA).
Phosphate buffered saline (PBS) (pH 7.4) was prepared by dilution from a previously prepared concentrated solution (×10) of PBS that contained KH2PO4 (17 mmol/L), Na2HPO4 (52 mmol/L), and NaCl (1.5 mol/L). Luria–Bertani media (LB) contained tryptone (10 g/L), yeast extract (5 g/L), and NaCl (10 g/L), pH 7.2 [28 ]. The LB + kan50 medium was prepared by the addition of a stock kanamycin solution (10 g/L) to a final concentration of 50 mg/L after autoclaving. Minimal salts medium (MSM) contained MgSO4·7H2O (0.1 g/L), NH4NO3 (0.2 g/L), trace elements (0.1 mL), 0.5 M phosphate buffer, pH 7.2 (100 mL), and distilled water (900 mL) [27 ].

Divinylbenzene/Carboxen/polydimethylsiloxane (DVB/CAR/PDMS) fibers and 20 mL SPME vials were purchased from Supelco (Bellefonte, PA, USA). All solvents were of analytical and HPLC-grade.
