Tocopherol and Phytosterol Quantification

Quantification of Tocopherol Content in Oils

The alpha-, gamma- and delta- tocopherol content of oil was determined by high-performance liquid chromatography (HPLC). To prepare samples for analyses, 2.5 g of oil were dissolved in 25 mL of HPLC-grade hexane. The tocopherol standards (Sigma-Aldrich, St. Louis, MO, USA) were also prepared at different concentrations (1, 2, 5, 10, 20, 50 and 100 ppm) in hexane. The samples and standards were filtered through 0.45 µm syringe filters into HPLC injection vials. The tocopherols were separated and quantified by injecting 50 µL of diluted standards and samples in a chromatograph equipped with an autosampler AS3000 (Oven temperature = 35 °C, tray temperature = 15 °C), a P1000 spectra system pump (Thermo separation products, San Jose, CA, USA), a Luna 5 µm normal phase column of 250 mm × 4.6 mm internal diameter (Phenomenex, Torrance, CA, USA) and an FP-2020 fluorescence detector (Jasco, Tokyo, Japan). The mobile phase used was 2% (v/v) tetrahydrofuran in n-heptane at a flow rate of 1.8 mL/min. The data were processed using the Chromquest 5.0 data processing software and the results are expressed as µg/g of oil.

Fatty Acid Profiling of Green Plum Seeds

Characterization of Raspberry Seed Oil

Quantification of Tocochromanols and Sterols

Quantifying Tocopherols in Liver Lipids

To examine tocopherol compounds in liver fat, the methodology by Ben Moumen et al. [31 (link)]. We employed high-performance liquid chromatography with diode array detection (HPLC-DAD) for this purpose. Initially, the samples were dissolved in hexane and then separated using an Uptisphere 120 A NH2 silica column (250 mm × 4.6 mm, 5 μm). The mobile phase used was a hexane/isopropanol mixture (99 : 1, v/v) maintained isocratically. The flow rate was set at 1 mL/min, and a UV detector was used to identify the separated compounds at wavelengths of 292, 296, and 298 nm. For quantification, we relied on external calibration, utilizing a commercially available tocopherol standard from Sigma-Aldrich (St. Louis, MO, USA).

Camellia Oils: Comprehensive Characterization

Seventeen camellia oils were collected from different factories in 2017. Among which, nine C. oleifera seed oils and three C. sinensis seed oils were purchased from the local market in Ganzhou city, Jiangxi Province of China; three C. japonica seed oils, two C. sinensis seed oils, extra virgin olive oil, and rapeseed oil were purchased from Tokyo in Japan. Detailed information describing the oil samples is shown in Table 1, which includes the brands, place of production, processing mode, and date of expiry. All the oil samples were stored at -20℃ in a freezer to prevent oxidation before analysis.
Boron trifluoride, tocopherol standard, lanosterol, squalene and 5α-cholestan-3β-ol were purchased from Sigma-Aldrich (St. Louis, MO, USA) . The other reagents were purchased from Wako Pure Chemical Co. (Osaka, Japan) . All chemicals were of analytical reagent grade, and acetone, acetonitrile, n-hexane and 2-propanol were HPLC grade.
