Oral Glucose Tolerance and Leptin

On day 4 after admission, a 75g oral glucose tolerance test (OGTT) was performed with glucose and insulin concentrations measured at −15, 0, 30, 60, 120 and 180 minutes. Glucose concentrations were measured using the glucose oxidase method (Beckman Glucose Analyzer 2, Beckman Instruments, Fullerton, CA, USA, or Analox GM9 Glucose Analyzer, Analox Instruments, Lunenburg, MA, USA). Glucose tolerance was classified according to American Diabetes Association criteria [21 (link)]. Plasma insulin concentrations were measured by AT AIA Pack IRI on TOSOH analyzer (Tosoh Bioscience, King of Prussia, PA, USA). Total and incremental area under the curve (AUC and iAUC, respectively) for insulin and glucose were calculated using the trapezoidal method. Plasma was available from 138 of the 196 subjects (70%) for leptin measurements at −15, 0, 30, 60, 120, and 180 minutes from the OGTT. Plasma leptin was measured in duplicate using ELISA kits (EMD-Millipore, Billerica, MA, USA). The intra-assay and inter-assay coefficients of variation (CV) for leptin was 5.3% and 9.2%, respectively.
