ABTS Assay for NOEO Antioxidant Activity

ABTS Radical Scavenging Activity Assay

The ABTS solution from the ABTS kit (S0119, Beyotime, Nantong, China) was mixed with the oxidant master mix in equal proportions and stored for 12 h before use, and 400 μL of the master mix was mixed with 13.6 mL of 0.01 M phosphate buffer (pH = 7.3, C0221A, Beyotime, Nantong, China) as the working master mix. Ten microliters of the sample solution containing different samples at a concentration of 0.5 mg/mL was added to 200 μL of working master batch and reacted for 5 min to measure at 734 nm using an ultraviolet-visible spectrophotometer (Infinite E Plex, TECAN, Männedorf, Switzerland). Trolox ((±)-6-Hydroxy-2,5,7,8-tetramethylchromane-2-carboxylic acid) was used to compare the ABTS radical scavenging activity with the TPs, and the results would be shown as mg Trolox equivalents (TE)/g TPs [20 (link)].

Rice Vinegar Characterization and Antioxidant Assays

Vinegar was provided by Jiangsu Hengshun Vinegar-Industry Co., Ltd (Jiangshu, China), which is one of the representative rice vinegars in China. Deionized water was prepared by a Millipore Milli-Q water system (Millipore, Bedford, MA, USA). Blue Dextran (MW: ~500 kDa), and Bromophenol blue (MW: ~670 Da), 2,2-azobis(2-amidinopropane) hydrochloride (AAPH), 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) and DCFH-DA ( ≥ 97% purity) were purchased from Sigma-Aldrich Co., Ltd. (Shanghai, China). ABTS kit was purchased from Beyotime Biotechnology Co., Ltd (Shanghai, China). DMEM, FBS, streptomycin, PBS and penicillin were purchased from Thermo Fisher Scientific (China) Co., Ltd (Shanghai, China). All the chemicals and solvents, unless otherwise noted, used were of AR grade, and Sepharose CL-4B column (60–2 × 104 kDa) were purchased from Sinopharm Chemical Reagent Co., Ltd (Shanghai, China).

Carnosol Antioxidant Activity Assay

Carnosol was dissolved in DMSO at a concentration of 40 mM for stock solution. Antioxidant activity detected by ABTS kit (Beyotime, Shanghai, China). The ABTS solution mixed with K2S2O8 to form a work solution and was stored at room temperature for 12 h–16 h before being used. Then the ABTS work solution was diluted with PBS for 40 times. 20 μL of carnosol at various concentration (10 ~ 0.04 μM) was mixed with 180 μL of diluted ABTS work solution. The absorbance at 734 nm was measured after incubation for 2–6 min at room temperature. We use the Absorbance (ODs) function of Flex station 3 (Molecular Devices, San Francisco, CA, USA) to detect the absorbance.
