HPLC Analysis of Amino Acids and Amines

DEEMM and L-ornithine monohydrochloride (≥99%) was obtained from Fluka. Boric acid (≥99%) was purchased from Hopkin & Williams. Amino acid commercial standards (L-alanine, L-arginine hydrochloride, L-asparagine, L-aspartic acid, L-glutamine, L-glutamic acid, L-glycine, L-histidine, L-isoleucine, L-leucine, L-lysine, L-methionine, L-phenylalanine, L-proline, L-serine, L-threonine, L-tryptophan, L-tyrosine and L-valine, ≥99.5%), γ-aminobutyric acid (≥99%), formic acid (≥95%), putrescine (≥99.5%), phenyethylamine (≥99.5%), tyramine hydrochloride (≥98%), ethylamine hydrochloride (98%), methylamine solution (40%), pyrrolidine (99%), ethanolamine (≥98%) and taurine (≥99%) were purchased from Sigma; dopamine hydrochloride (99%), histamine dihydrochloride (≥98%) were purchased from Alfa Aesar; L(+)-α-aminobutyric acid (98%) were purchased from Acros Organics; β-aminobutyric acid (98%) were purchased from BLD Pharmatec. Ultrapure water was supplied by a Millipore Milli-Q Advantage A10 (Millipore) and 0.75 M borate buffer at pH = 9 was prepared in deionized water.

Inflammatory Pain Modulation Protocols

L-arginine hydrochloride (NO synthase (NOS) substrate), L-NAME (non-selective inhibitor of NO synthase), and diazoxide (an ATPsensitive K + channel opener) were obtained from Sigma-Aldrich (St. Louis, Missouri, USA). Methylene blue (NOS and guanylyl cyclase inhibitor) was provided by Merck. Glibenclamide (ATP-sensitive K + channel inhibitor) and carbamazepine were kindly donated by Darupakhsh and Abidi Pharmaceutical Co. (Tehran, Iran), respectively. Pioglitazone (a PPARγ agonist) and GW-9662 (a PPARγ antagonist) were purchased from Osveh Pharmaceutical Co, and Tocris Bioscience, respectively. All drugs were dissolved or suspended in normal saline (0.9 % NaCl) and buffered to a pH of 7.3. Drug concentrations were freshly prepared in such a way that the necessary dose could be injected in a volume of 50 µL/paw by the intraplantar (i.pl.) route. Doses and drug administration schedules (▶Fig. 1) were selected based on previous reports and also our experience in the laboratory [7, 11, 17, 20] .

Investigating HSF1 and SIRT1 Regulation

In situ cell death detection kit was purchased from Roche Applied Sciences (Mannheim, Germany). PureProteome™ Protein A and Protein G magnetic beads were purchased from Merck Millipore (Billerica, MA, USA). Other reagents were analytically pure.

Induction and Assessment of Severe Acute Pancreatitis

SAP was induced by two intraperitoneal injections of L-arginine hydrochloride (Sigma-Aldrich, St. Louis, MO, USA) (8%, pH=7.0), 4 g/kg bodyweight each, with 1-hour interval between injections as previously reported.15 (link) Sham injections of NaCl were administrated to normal controls. Mice were then injected with phosphate-buffered saline (PBS) or recombinant murine IL-22 (rIL-22; 200 ng/mouse, Miltenyi Biotec, Bergisch Gladbach, Germany) at specified times. All mice were returned to the cages, with free access to food and water. SAP group mice were sacrificed at 24, 48, and 72 hours after the second injection of L-arginine, respectively.

SILAC Cell Culture Labeling Protocol

Mouse Models of Pancreatitis Induction

The protocol for animal use was reviewed and approved by the University of Pittsburgh Institutional Animal Care and Use Committee. RAGE−/− mice (SVEV129×C57BL/6) were a kind gift from the late Dr. Angelika Bierhaus (12 (link)). TLR4−/− mice (C57BL/6 background) and AIM2−/− mice (C57BL/6 background) were obtained from The Jackson Laboratory (Farmington, CT, USA). For L-arginine-induced pancreatitis, a sterile solution of L-arginine hydrochloride (8%; Sigma) was prepared in normal saline and the pH was adjusted to 7.0. Mice received three hourly intraperitoneal (i.p.) injections of L-arginine (3 g/kg), while controls were administered saline i.p. as a control as described previously (13 (link)). For cerulein-induced pancreatitis, mice received seven hourly i.p. injections of 50 μg/kg cerulein (Sigma) in sterile saline, while controls were given saline as described previously (14 (link)).
