Evaluating Bevacizumab Effects on mIDH1-U87 Cells

The mIDH1-U87 (U87 cell line stably transfected with IDH1-R132H with a lentiviral vector, and was kindly provided by Marc Sanson [Hôpital de la Salpêtrière, Paris, France]) was maintained in Eagle’s minimal essential medium (EMEM) with 10% foetal calf serum, 2 mM L-glutamine, 100 U/mL penicillin, and 100 μg/mL streptomycin (Lonza, Verviers, Belgium). bevacizumab (Roche, Paris, France) was diluted with culture medium to working concentrations before use. As a control, a stock solution containing the corresponding excipient was prepared with 60 mg/mL of trehalose dehydrate, 5.8 mg/mL sodium dihydrogen phosphate monohydrate, and 1.5 mg di-sodium hydrogen phosphate dihydrate (all from Sigma Aldrich, Saint-Quentin Fallavier, France). VEGF secretion was assessed with the Quantikine ELISA kit for human VEGF (R&D Systems, Abingdon, UK), following the manufacturer’s instructions. The cells proliferation was assessed as follows: The mIDH1-U87 cells were seeded into and allowed to attach overnight. Cells (seeded in 24-well plates; 30,000 cells/well) were treated for 72 h with bevacizumab (from 0.1 mg/mL to 1 mg/mL) in triplicate wells. Duplicate experiments were performed. Cell viability was then assessed with the MTT assay following the procedure of Mosmann.13 (link) The percentage of surviving cells is expressed as the ratio of optical density of treated cells versus untreated cells.

Preparation of Phosphate Buffer in D2O

Deuterium oxide 99.9%, sodium dihydrogenphosphate hydrate (NaH2PO4·H2O), disodiumhydrogen phosphate dihydrate (Na2HPO4·H2O2), and 3-(trimethylsilyl)propionic-2,2,3,3-d4 acid sodium salt (TSP) were purchased from Sigma-Aldrich (Milan, Italy). Deuterated EDTA d-16 98% was obtained from Product Cambridge Isotope Laboratories, Inc.
Phosphate buffer solution was prepared as follows: 49.7 mg of sodium dihydrogenphosphate hydrate (0.36 mmol), 202.9 mg of disodium hydrogen phosphate dihydrate (1.14 mmol), and 0.92 mg deuterated EDTA d-16 (0.003 mmol) were dissolved in 10 mL of water. The pH was checked at 7.1. The solution was distributed into 5 mL aliquots and then lyophilized. Each aliquot was dissolved in about 1 mL of D2O and lyophilized again. Finally, the buffer was dissolved in 5 mL of deuterium oxide with 0.002% TSP (12 mM).

Acetylthiocholine iodide (ATCh), DTNB (5,5'-dithiobis(2-nitrobenzoic acid)), BW284C51 (1,5-bis(4-allyldimethyl-ammoniumphenyl)-pentane-3-one dibromide), ethopropazine ((10-[2-diethylaminopropyl] phenothiazine)hydrochloride), and chemicals for high performance liquid chromatography (HPLC) [acetonitrile gradient grade LiChrosolv, disodium hydrogen phosphate dihydrate, citric acid monohydrate, ethylenedinitrilo tetraacetic acid disodium salt dihydrate (Na 2 EDTA), 1-octane sulphonic acid sodium salt, perchloric acid (70 %), and phosphoric acid] were purchased from Sigma-Aldrich (St. Louis, MO, USA). The water used for HPLC was double-distilled and deionised HPLC-grade.
