Bioactive Compound Extraction and Evaluation

Ascorbic acid, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS●+), neocuproine, trichloroacetic acid (TCA), and potassium ferricyanide were used for the determination of the antioxidant activities and were purchased from Sigma Chemical Co. (Sigma-Aldrich GmbH, Stern-heim, Germany). The following chemicals were used for the enzyme inhibition assays: α-glucosidase from Saccharomyces cerevisiae, 4-nitrophenyl α-D-glucopyranoside (pNPG), α-amylase from porcine pancreas, β-nicotinamide adenine dinucleotide (β-NADH), phenazinemethosulphate (PMS), nitrotetrazolium blue chloride (NBT) and Ascorbic acid were obtained from Sigma (St. Louis, MO, USA), while acarbose and potato starch were purchased from Fluka (Bucharest, Romania) and Fisher (Pittsburgh, PA, USA), respectively. Sodium nitroprusside, sulfanilamide, and 3,5-dinitrosalicylic acid (DNS) were obtained from Acros Organics (Hampton, NH, USA). Folin–Ciocalteu reagent, Na2CO3, and gallic acid were purchased from Panreac (Barcelona, Spain). Standards used for the elucidation of the phenolic compounds and for elaboration of the calibration curves were obtained from EXTRA SYNTHESE (Genay Cedex, France). Acetonitrile HPLC-grade and formic acid were purchased from Panreac (Barcelona, Spain). All other chemicals were of analytical grade.

Quantification of NAD+ via HPLC

Comprehensive Analytical Protocol for Natural Antioxidants

Hydroxytyrosol (98% of purity, w/w) was procured from Extrasynthese (Lyon, France). Ultrapure water was obtained using a Milli-Q system (Millipore, Bedford, MA, USA).

Characterization of Synthetic Matrices

Bovine Hemin Porphyrin Extraction

Extraction and Formulation of Jatrophone

The dichloromethane fraction from J. isabellei (DFJi) was resuspended in a mixture composed of dimethyl sulphoxide (DMSO), polyethylene glycol 400 (PEG 400), and phosphate buffered saline (PBS), pH 7.4 (5:47.5:47.5 v/v) for oral administration (p.o.) in rats. For intravenous administration, the fraction was dissolved in a mixture composed of DMSO, polysorbate 80, and saline solution (5:4:91 v/v). A combination of κ- and λ-carrageenan was purchased from BDH Chemicals Ltd. (London, UK). Dexamethasone and indomethacin were purchased from Deg (São Paulo, Brazil), and colchicine from Sigma-Aldrich (St Louis, MO). The jatrophone isolation from the dichloromethane fraction required the use of Silica Gel 60 and Silica Gel 60 F254-coated plates, which were purchased from Merck (Darmstadt, Germany). Analytical grades hexane, acetone, dichloromethane, ethanol and methanol were purchased from Vetec (Duque de Caxias, Brazil). Acetonitrile HPLC grade was purchased from Panreac (Barcelona, Spain). The ultrapure water utilized in the UFLC analyses was obtained using a Milli-Q purification system (Millipore, Billerica, MA).

The lutein epoxide, luteoxanthin, zeinoxanthin, 9-cis-antheraxanthin, 9-cis-violaxanthin, 13-cis-violaxanthin, and 9-cis-lutein were obtained as described elsewhere [23 (link),24 (link),25 (link),26 (link),27 (link)]. The gallic acid, p-hydroxybenzoic acid, syringic acid, caffeic acid, m-coumaric, p-coumaric, chlorogenic acid, ferulic acid, naringin, naringenin, ethyl galate, quercetin, kaempferol, crisin, vanillic acid, and myricetin were purchased from Sigma-Aldrich (Madrid, Spain). The quercitrin was obtained from Extrasynthese (Genay, France). All the aqueous solutions were prepared with purified water in a NANOpure DlamondTM system (Barnsted Inc., Dubuque, IO, USA).
