Multiparametric Flow Cytometry Analysis

Single-cell suspensions from BM and spleen were treated with Gey’s solution to remove red blood cells and resuspended in PBS supplemented with 2% BSA. The cells were then stained with a combination of fluorescence-conjugated antibodies. APC-conjugated anti-B220, anti-IgM, anti-CD4 and anti-CD44, and PE-Cy7-conjugated anti-B220, anti-IFNγ, anti-CD25 and anti-CD23 were purchased from eBioscience. PE-conjugated anti-CD43, anti-CD21, anti-Thy1.2, anti-CD5, anti-CD8, and anti-IgD were purchased from BD Biosciences. Samples were applied to a flow cytometer (LSRII, Becton Dickinson). Data were collected and analyzed using FACSDiva software (Becton Dickinson) or FlowJo software (Tree Star).

Multimodal Immune Profiling of Tissue

Th17 (CD4, IL-17), Treg (CD4, CD25, Foxp3), SMILE, AMPK, mTOR, pSTAT3 705, pSTAT3 727 and germinal center markers (CD3, B220, PNA) expression levels were analyzed by confocal microscopy. Tissue Sects. (7 μm thick) were fixed in methanol-acetone and stained with phycoerythrin (PE)-conjugated anti-CD4 (Biolegend, San Diego, CA, USA), fluorescein isothiocyanate (FITC)-conjugated anti CD4 (BD Bioscience, San Diego, CA, USA), PE-conjugated anti-interleukin-17 (eBioscience, San Diego, CA, USA), allophycocyanin (APC)-conjugated anti-CD25 (Biolegend), PE-conjugated anti-Foxp3 (Thermo Fisher Scientific, Waltham, MA, USA), PE-conjugated anti-phosphorylated Stat3 (Tyr 705, Ser 727, BD Bioscience), APC-conjugated anti-B220 (eBioscience), Alexa 594-conjugated anti-PNA (Thermo Fisher Scientific), SMILE (Abcam, Cambridge, UK), AMPK (Abcam) and mTOR (Cell Signaling). After overnight incubation at 4℃, the stained sections were analyzed on a Zeiss confocal microscope (LSM 700; Carl Zeiss, Oberkochen, Germany) at 200× magnification and Zen Blue edition software.

Psoriasis Induction and Analysis Protocol

Fluorescein isothiocyanate (FITC)conjugated anti-CD3, phycoerythrin (PE)-conjugated anti-CD4, PE-conjugated anti-CD11c, allophycocyanin (APC)-conjugated anti-CD11b, FITC-conjugated anti-F4/80, APC-conjugated anti-B220, PE-conjugated anti-langerin, APC-conjugated anti-EpCAM all were from eBioscience, Darmstadt, Germany, while peridinin chlorophyll protein complex (PerCP/Cy5)-conjugated anti-Siglec-H was from Biolegend, Koblenz, Germany and PerCP-conjugated anti-CD8 was purchased from BD Bioscience, Heidelberg Germany.
