Intracellular Cytokine Staining of T Cells

Intracellular cytokine staining was performed as previously described (Lamoreaux et al., 2006 (link)). Briefly, TILs or PBLs were activated with 50 ng/mL PMA, 1 μg/mL ionomycin (EMD Chemicals) or DMSO for 2, 4, 8 h, or with peptide (10 ug/mL) or 8 h, and monensin (BD Biosciences) was added for the last 2–8 h of incubation. Treated cells were then fixed and permeabilized according to manufacturer’s protocol (BD Biosciences, 554715). Staining was performed with FITC anti-human IFN-γ antibody (Biolegend, 502507).
