The Cell Counting Kit‐8 Reagent is a colorimetric assay for the determination of the number of viable cells in cell proliferation and cytotoxicity assays. The reagent contains a water‐soluble tetrazolium salt, which is reduced by dehydrogenases in living cells to produce a colored formazan dye. The amount of the formazan dye is directly proportional to the number of living cells, and can be measured by a spectrophotometer.

Optimizing bEnd.3 Cell Viability

bEnd.3 cells were treated with different concentrations of H2O2 (0, 500, 700, 800, 900 and 1000 µm) and NMN (0, 100, 200, 300, 500 and 600 µm) for 24 h. Cell Proliferation Assay Protocol

A total of 3 × 103 cells were inoculated in a 96-well plate. After inoculation for 24 h/48 h/72 h/96 h, 10 μL of Cell Counting Kit-8 reagent (CCK-8; Abmole, Shanghai, China) was added to each well and incubated for 2 h at 37°C. The absorbance of each well was determined at 450 nm.
