Sch B Induces Apoptosis in Cancer Cells

Sch B and Rhodamine 123 (Rho 123) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Sch B was dissolved in dimethyl sulfoxide (DMSO; Sigma-Aldrich) to create a 100 mM stock solution. The stock solution was further diluted with culture media to acquire the desired concentrations. The control groups were treated with equal volumes of DMSO. Cell Counting Kit-8 (CCK-8) was purchased from Dojindo Laboratories (Tokyo, Japan). The Annexin V/PI apoptosis kit was obtained from BD Biosciences (San Diego, CA, USA). Acquisition of Analytical-Grade Compounds

Salmonella-Mediated Cancer Therapy

Salmonella enterica serovar choleraesuis (S. Choleraesuis; S.C.) (ATCC 15480) was obtained from Bioresources Collection and Research Center (Hsinchu, Taiwan) 10 (link). B16F10 (mouse melanoma) 11 (link) and 4T1 (mouse breast cancer) 12 (link) cells were maintained in culture dish with Dulbecco's Modified Eagle Medium containing 1% antibiotics (100 units/mL penicillin and 100 μg/mL streptomycin), 2mM l-glutamine and 10% fetal bovine serum. Rhodamine 123 (Rho-123) and 5-Fluorouracil (5-FU) were purchased from Sigma Aldrich. Constitutively active AKT plasmid was kindly provided by Dr. Chiau-Yuang Tsai (Department of molecular immunology, Osaka University) 13 (link). Cells were treated with plasmids for 16 h in serum free medium prior to adding Salmonella (multiplicity of infection (MOI) = 10) into cells for 90 min. C57BL/6 (B16F10 mouse tumor model) and BALB/c (4T1 mouse tumor model) female mice were purchased from the National Laboratory Animal Center of Taiwan. The animals were maintained in a pathogen-free animal care facility in isothermal conditions with regular photoperiods. The experimental protocol adhered to the rules of the Animal Protection Act of Taiwan and was approved by the Laboratory Animal Care and Use Committee of the National Sun Yat-sen University.

Rhodamine-123 Uptake Assay for BMDMs

BMDMs or Abcb1b–/– clones were seeded into 96-well plates at a concentration of 1 × 105 cells per well. Where indicated, BMDMs were treated with elacridar overnight (2–10 μM). Rhodamine-123 (Rho123; 2 μM, Sigma) was added, and cells were incubated at 37° C for 30 min. Cells were then washed with PBS three times to remove any extracellular Rho123, and DMEM were replaced, after which cells were incubated for a further 30 min at 37° C. Cells were then washed again with PBS and lysed in cell lysis buffer before being read at excitation and emission wavelengths of 485 and 535 nm, respectively, in a fluorescent plate reader.

Isolation and Characterization of Rat Notochordal Progenitor Cells

The anti-inducible nitric oxide synthase rabbit polyclonal (iNOS; sc-651) antibody was from Santa Cruz Biotechnology, Inc. (Dallas, TX, USA). Rhodamine 123 (Rho123) and Hoechst 33342 were obtained from Sigma-Aldrich (St. Louis, MO, USA). Phosphate-buffered saline (PBS, ×10, ST476) and SDS-PAGE Gel Kit (P0012A), were obtained from the Beyotime Institute of Biotechnology (Haimen, China) and were of the highest purity commercially available.

Cytotoxicity Evaluation of Sesquiterpenes
