Regorafenib Treatment on Cardiomyocytes

Check if the same lab product or an alternative is used in the 5 most similar protocols
AC16 human cardiomyocyte cell line was purchased from American Type Culture Collection (ATCC, Rockville, MD). Under conditions of CO2 incubators maintaining atmospheric oxygen levels and 5% CO2, cardiomyocyte cells were cultured using Dulbecco's modified Eagle's medium (DMEM, Gibco, 11965092, USA) with 10% fetal bovine serum (FBS, Gibco, 16140071, USA). Cells were passed regularly and subcultured to 80% confluence before the cell experiment. For 12 h prior to each experiment, the cell medium was supplemented with 0.5% FBS. A concentration of 1 μM of regorafenib (HY-10331, New Jersey, USA) was then applied to the plates for 48 hours. Regorafenib was melted in dimethyl sulfoxide (DMSO, Biosharp, BS087, China), and DMSO was used as a control at a concentration of 0.1 nM.

Proliferation Analysis of lncRNA STEAP3-AS1

Check if the same lab product or an alternative is used in the 5 most similar protocols
The proliferation of lncRNA STEAP3-AS1 was examined by methyl thiazolyl tetrazolium (MTT) (Sigma, USA) assay. Briefly, cells were cultured in 96-well plates at a density of 5 × 103 cells per well for 24, 48, and 72 h. Then, 20 μL of MTT reagent (5 mg/mL) was added to each well and incubated at 37°C and 5% CO2 in a humidified incubator for 4 h, followed by removing medium and adding 200 μL of dimethyl sulfoxide (BioSharp, China) to each well and shaken on a rotary platform for 10 min to dissolve formazan crystals. A microplate spectrophotometer (Bio-Rad, USA) was used to measure the optical density of each well at 570 nm.

MTT Assay for Fibroblast Viability

Check if the same lab product or an alternative is used in the 5 most similar protocols
Fibroblasts at a density of 1.5×105 cells/ml in 200 μl were seeded into a 96-well plate. Following addition of 20 μl MTT solution (5 mg/ml; Beyotime Institute of Biotechnology, Jiangsu, China), plates were incubated in a humidified environment with 5% CO2 at 37°C for 4 h. Dimethyl sulfoxide (200 μl; Biosharp) was added to each well following supernatant removal to dissolve the purple crystals. Absorbance was then read at 490 nm on a microplate reader (Thermo Fisher Scientific) and the inhibition rate (IR) in each experimental group was calculated as follows: IR (%)=[(ODcontrol group-ODexperimental group)/ODcontrol group]x100%.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
