The GCaMP loci were imaged with a doubler in addition to Olympus LUMPlanFL lens 40x/0.80 N.A. water immersion objective through the bandpass filters (λEx: 490–510 nm, λEm: 520–550 nm) for quantitatively measuring the effect of transient ionomycin puffing on GCaMP fluorescence.
