Investigating APP Overexpression in Trophoblast Cells

The functional implication of APP overexpression was investigated in a previously established trophoblast cell line HTR-8/SVneo. 17 HTR-8/SVneo was stably transfected with a Tet-On expression plasmid harboring APP ORF pTRE3G-BI-APP. Two clones, B2 and B10, were selected. APP was induced in B2 and B10 by including the recombinant protein Tet-Express (Clontech) in the culture medium.
For assessing the impact of APP overexpression on syncytialization, BeWo was first stably transfected with pCMV-TET3G, which expresses the Tet-On 3G transactivator protein. BeWo-tet3G was then transiently transfected with pTRE3G-BI-APP using PolyJet (Signagen, Rockville, MD), according to the manufacturer's protocol, and APP was induced with doxycycline.
