Zucker Diabetic Fatty Rat RYGB Study

Male Zucker Diabetic Fatty (ZDF) rats (Charles River Laboratories, Wilmington, MA), homozygous recessive for the leptin receptor (fa/fa), were randomized at age 12 weeks to RYGB (n=16), sham-operated (SHAM, n=16), or pair-fed (PF, n=16) groups. After intervention, the rats were followed for 30 days. The PF group did not undergo laparotomy and were fed the average daily intake of the rats that underwent RYGB. All rats had ad libitum access to water and the diabetogenic Purina 5008 diet with 56.4% carbohydrate (Research Diets, New Brunswick, NJ). Rats were housed in individual cages in the Cleveland Clinic Biological Resources Unit with temperature controlled to 22°C and a 12h-dark/light cycle. All animal experimental procedures were approved by the Cleveland Clinic Institutional Animal Care and Use Committee. The National Institute of Health Animal Use Guidelines were adhered to for this study. All efforts were made to minimize any suffering.

Animal Care Protocol for Metabolic Disorders

All experimental procedures were approved by the institutional animal care and use committee of Mitsubishi Tanabe Pharma Corporation (Osaka, Japan) and animal care was conducted in accordance with institutional guidelines. Male KK-Ay mice were purchased from CLEA Japan (Tokyo, Japan), and male Zucker diabetic fatty (ZDF) rats and Zucker lean (ZL) rats were purchased from Charles River Laboratories Japan (Tokyo, Japan). The animals were individually housed in plastic cages with ad libitum access to standard chow (CRF-1; Oriental Yeast Co., Ltd., Tokyo, Japan) and tap water. The animal rooms were controlled for temperature (23 ± 3°C), humidity (55 ± 20%), and a 12-h light/dark cycle. These animals were acclimated for 1 week prior to the start of the studies.

All animal handling and experimental procedures were approved by the Institutional Animal Care and Use Committee at the University of North Carolina – Chapel Hill. (Ref no. IACUC16-254 (10-2016-09-2019)). Male Zucker Diabetic Fatty (ZDF) rats were obtained from Charles River Laboratories (Wilmington, MA). Animals were fed Irradiated Purina 5008 (Granville Milling, Granville, OH) chow. Blood glucose was measured by tail nick using a Freestyle Precision Neo Blood Glucose Monitoring System (Abbott Laboratories, Abbott Park, Illinois). Glycemia > 400 mg/dL after two consecutive random weekly measurements was used to determine type 2 diabetes onset.
