Synthesis of Functional Polymer Nanoparticles

The monomers used were 2-(dimethylamino) ethyl methacrylate (DMAEMA) (98%), lauryl methacrylate (LMA) (96%) and (oligo ethylene glycol) methacrylate (OEGMA, average Mn = 475) from Sigma-Aldrich (St. Louis, MO, USA). They were purified by passing through an inhibitor removing mini column. Resins used were 311,340 and 311,332 from Sigma-Aldrich. The 2,2-azobis(isobutyronitrile) (AIBN) was the radical initiator utilized after recrystallization from methanol. The 1,4-dioxane (99.8% pure), from Aldrich, was dried over molecular sieves before use. Tetrahydrofuran (THF, 99.9% pure), 4-cyano-4-(phenylcarbonothioylthio) pentanoic acid (CPAD), fetal bovine serum (FBS) and other reagents from Aldrich were used as received.

Potassium ethyl xanthogenate (96%), 2-bromoacetic acid, 4-(dimethylamino) pyridine (DMAP, >99%), N,N′-dicyclohexylcarbodiimide (DCC, >99%) and 2-bromoacetic acid were all purchased from Alfa Aesar and used as received. 4-(Dimethylamino)pyridinium-4-toluene sulfonate (DPTS) was synthesised as previously reported.25 (link) Polyacrylic acid (Carbopol 940) was purchased from Lubrizol. All solvents, unless stated otherwise, were reagent grade, purchased from Fisher Chemicals and used as received.

Synthesis of Clickable Biomimetic Polymers

Polycaprolactone (M n = 80 kg mol À1 ), oligoethylene glycol methacrylate (OEGMA) with an average molecular weight of 475 (methyl ether termination), 2,2 0 -bipyridine, CuBr, NaCl, NaH 2 PO 4 and Na 2 HPO 4 were obtained from Sigma Aldrich. Prior to use, CuBr was purified by washing three times with glacial acetic acid, twice with absolute ethanol and twice with diethyl ether followed by drying in a vacuum oven overnight. KF (anh.), Na 2 EDTA, CuSO 4 and ascorbic acid were acquired from BDH. Fetal bovine serum was purchased from Life Technologies. The protected alkyne monomer, TMS-propargyl-hexaethylene glycol methacrylate (TMS-Prg-HEGMA), 40 the click reaction ligand, tris((1-(4-carboxybenzyl)-1H-1,2,3-triazol-4-yl)methyl)amine) (TBTA-COOH), 41 and the fluorinated azide labelling molecule, 2,2,2-trifluoroethyl 4-(azidomethyl)benzoate (TFAB), 31 were synthesised as previously reported. A cyclic RGD peptide, cyclo(Arg-Gly-Asp-D-Phe-Lys(azide)) was obtained from Peptides International (Louisville, KY, USA). All solvents (chloroform, methanol, ethanol (96%), DMF, DMSO and toluene) were reagent grade unless specified and obtained from Merck. All water was purified and deionised using a MilliQ system (Millipore).
